Prostate cancer is the leading malignant tumor in genitourinary system of old men. It is one of the most common cancers in the western nations .With the population ageing and the changing of food and drink and life style, prostate cancer is increasing quickly .Copper and Zinc is the important moity of metal enzyme, which have kinds of physiologic functions in vivo. The content of Zinc in normal prostate tissue is higher than that in any other tissues in Zinc human body, about 3 to 10 fold greater than that of others . But the content of Zinc in prostate cancer tissue is considerably lower than that in normal prostate tissues. Copper may cause cancerogenesis, excessive copper contribute to DNA and induce cell malignant differentiation and proliferation.It increases free radical, cause lipid peroxidation, results in biological damage and cell cancerization.Prostate specific antigen is the most important tumor marker at present, it has height organ specificity. PSA is the best index in the screening of prostate cancer, auxiliary diagnosis and monitoring therapeutic effectMatrix metalloproteinase is one kind of ecto-cell proteolytic enzyme with dependent of Zinc can degrdn basal membrane, promote invasion and metastasis of malignant tumour cells. Past research indicates that matrix metalloproteinases-2 and matrix metalloproteinases-9 intimately relation to prostate carcinogenesis,invasion and metastasis of prostate cancer.Objective:(1) Study the relationships between changes of serum Zinc, Copper and prostate cancer about people prostate cancer clique extensive survey. Comparing with efficiency in diagnose of prostate cancer with serum prostate specific antigen. .(2) Investigate the effect of invariably dose Zinc on the expression of MMP-2 andMMP-9 mRNA of mouse RM-1 in prostate cancer.Method:(1)Use Hitachi 180-80-type polarize Zeeman effect atomic absorption spectrophotometer determine the value of serum Copper and serum Zinc.(2) Apply ELISA method, determine serum PSA.(3) MTT experiment can detect the effect of diversity dose Zinc on RM-1 cells multiplication.(4) Contrast phase micro observe corpuscular morph change of Zinc on RM-1 cell.(5) RT-PCR detect the effect of invariably dose Zinc on the expression of MMP-2 andMMP-9 mRNA of mouse RM-1 in prostate cancer.Result:(1)Serum zinc of prostate cancer group is obviously higher than healthy control group,serum Copper and ratio of Cu/Zn is obviously super to healthy control group(P<0.05).Serum Copper and ratio of Cu/Zn between prostate cancer group and hyperplasia of prostate gland group ,there is unknown significant difference; Serum Zinc of prostate cancer group is obviously lower than hyperplasia of prostate gland group(P<0.05).There is unknown significant difference between Serum Zinc of hyperplasia of prostate gland group and Serum Zinc of healthy control group ;Serum Copper and ratio of Cu/Zn in hyperplasia of prostate gland group is obviously super to healthy control group(P<0.05).(2) When PSA exceed 4.0ng/ml,area under curve of PSA is Percentage of seventyseven point six, which is super to area under curve of serum Zinc, Copper. When PSA in 4.0-10.0ng/ml grey region , The percentage of region under curve of serum Zinc is sixty six ,which is super to area under curve of PSA whose percentage is sixty one point nine, therefore when PSA in 4.0-10.0ng/ml grey region efficiency of diagnosis prostate cancer is super to PSA.(3) MTT experimental result indicate that when the density of Zn2+ is in 3.125-100μM, the survival rate of RM-1 cells is slightly super to healthy control group;when the density of Zn2+ is in 100-400μM, the survival rate of RM-1 cells is obviously infra to healthy control group(P<0.05).(4) Contrast phase micro observe change of corpuscular morphology after Zinc effect on RM-1 cells, when the density of Zn2+ reaches 200μM, morphologic changes of cell more obviouse,cells change long ,and stretch out longer ecptoma., phasing of cell division decreases. obviously cell population decreases. Obviously compare to that of control group. When density of Zn2+ reaches 400μM, RM-1 cell bulk miss origin cells morph cells shrink and change round.and float in culture fluid..(5)RT-PCR result in display the density of Zn2+(150μM,200μM,250μM)effect on RM-1 cells after 24 hours ,expression quantity of MMP-2.andMMP-9 mRNA attenuate obviously compare with that of control group.Conclusion:(1)Finding of crowd of prostate cancer indicates that the serum Zinc of prostate cancer is infra to healthy group and hyperplasia of prostate gland group.(2)Serum Zinc conduce to elevate efficiency of diagnosis prostate cancer when PSA in 4.0~10.0ng/ml gray area.(3)Extraorgan essay indicate that invariably dose Zinc can inhibit expression of MMP-2 andMMP-9 mRNA of mouse RM-1 in prostate cancer,which inhibit invasionand extension of cancer.Finding indicates that the depression of serum zinc has dependability with prostate carcinogenesis,Zinc may inhibit invasion and extension of cancer.in some degree.
