ObjectiveTo discuss the effects of acrylamide (AA) on the content of procreation in male rats and measure the content of T,FSH,LH ; NO,NOS;activity, quantity and morphology of sperms;and cell cycle; proliferation in testicle tissue in rats after sub-chronic toxicosis. At the same time in search of forepart target of it's toxicity and discuss maginabale mechanism of procreation toxicity.MethodsThe male mice were randomly divided into negative control group , AA (low dose, middle dose and high dose) groups, receiving normal saline, AA(5,10,20 mg/ kg)respectively by gavage once a day, five times per week. The mice were sacrificed after eight weeks form the first gavage,took blood by methold of vein and separated serum by automatic to check it's T,FSH,LH,measured the content of NO ,NOS in testicle tissue and the weight of testis,took one side of epididymis to take sperm's count,activity and morphology .At the same time,we can observe the change of testis by pathology .Results1. Measure of weight: the weight of control group ,low-dose group and media dose group were increased by installing fluid into stomach, especially control group was increased more obvious, low-dose group was second ,and media-dose group was the last one,but high-dose group'weight was decreased.2. Testis and sperm: The weight of testis ,sperm's count and activity was obvious decreased with dose increasing , and each dose group was difference comparing to control group(p<0.01). The sperm deformation rate was also increased with dose increasing, and there was obvious difference between each dose group and control group(p<0.01) but low dose.3. Measure'results of NO, NOS in testis tissue: NO, NOS in testis were a little decreased with dose increasing , the NO of each dose group was difference comparing to control group (p<0.05),the NOS of high dose group was difference comparing to conterol group (p<0.05).4. T,FSH,LH's content in serum: T and LH's content in serum was decreased with dose increading, and there was obvious difference in each dose group comparing(p<0.01). FSH's content there was not obvious difference in each dose group comparing.5. The cell cycle and languish: The cell cycle was foul-up,the cell languish was decreased with dose increading,and middle dose and high dose were difference comparing to control group(p<0.05) .6. Pathology of testis tissue: We can observe partly cell's arragment disorder in testis tissue, and separation between upper and base part, but there was no blood or necrosis in testis. ConclusionThere is obvious toxicity of ACR on male mouse ,and the quantityof sperm and activity is decreaed , the rate of deformation is increased. T and LH's content in serum were decreased, and the specific enzyms of NO, NOS in dose testis tissue are difference comparing to control group. The cell cycle was foul-up,the cell languish was decreased with dose increading.
