Follistatin (FS) is a single polypeptide chain,which was isolated from ovarian follicular fluid and directly inhibit the secretion of FSH from anterior pituitary cells. Follistatin has a widely histological distribution, follistatin mRNA with highest expression in adult ovary,pituitary,kidney,in fetal heart and liver. The character of FS expression indicates that it may have a different function on different kind of cells. FS can specifically binds albumen and activin, directly inhibit biological activity and take part in many adjusting of physical function. Mouse gene knock indicate: FS plays important role from former of embryo to maturity of organism. FS often coexist and express with activin to make a balance system for maintaining normal growth of organization and organ. One is over expression or another decrease will result in imbalance of FS-ACT system, and with physiological defect and the course of pathology of some organization and organ. In the present study, FS was highly involved in forming ofhepatic fibrosis, lung fibrosis, renal injure, injure of skin, injure of brain, atherosclerosis, PCOS, disease of reproduction and gonad.1. Research objectiveTo investigate FS`s expression and clinical application in placental trophoblast cell,possible origin of peripheral blood FS in pregnancy phase, secretive pathway and influential factor.2. Research methods(1) The peripheral blood circular FS mRNA levels of different pregnancy phase and the normal health group was tested by using fluorescent-quantitative RT-PCR.(2) Placental tissues in pregnancy are analyzed by immunohistochemistry staining.(3) To observe the origin of peripheral blood FS in pregnancy, we selected placental tissue with high peripheral blood FS, separated placental trophoblast cell and cultivate it, FS levels was examined in ectogenetic pregnant cultivated liquid by ELISA.3. Research results(1) The levels change in peripheral blood circular FS mRNA in early pregnant is no more than that of the no-pregnant normal health group and late pregnancy. There is no difference by statistics evidently. P>0.05.The levels change in peripheral blood circular FS mRNA in overdue pregnancy is lower than that of mature pregnancy . there is yet no difference by statistics evidently, P>0.05. The levels change in peripheral blood circular FS mRNA in threatened abortionpatient rise higher than that of the early, there is no difference evidently between them. p<0.05. It is yet hard to avoid the levels of threatened abortion patient change lower significantly. Difference is obvious to compare to the early, also to be by statistics. p<0.05 .the test to expression levels of placental chorionic histiocyte by PCR indicate that the early placental chorionic histiocyte expression levels is higher than the late, there is difference obvious, p<0.05.The FS mRNA expression levels of threatened abortion patient chorionic histiocyte rise higher than that of the early, there is difference between them obviously, p<0.05.it is yet hard to avoid The FSmRNA expression levels of threatened abortion patient chorionic histiocyte change lower significantly. Difference is obvious to compare to the early. p<0.01.(2) Immunohistochemistry showed that the early pregnancy placental chorionic organization anti-FS antibody group staining is strong, that the late pregnancy placental chorionic organization anti-FS antibody group staining yet is weak.(3) The increase of peripheral blood FS in pregnancy phase is probable directly rooted in secretion by placental trophoblast cell.4. Research conclusionPlacental organization can produce FS, FS expression levels is related to pregnancy physiology and pathology.
