| Rheumatoid arthritis(RA), a common and severe disease, is a chronic inflammatory disease of unknown cause, which is characterized membrana synovialis hyperplasia, inflammation and joint destruction. Its prevalence in adults in China is about 0.32～0.35﹪, which is lower than that in America 1﹪. And it is one of major diseases causing disability in our nation. The major pathological change of RA is synovitis, with the invasion of cartilage and joint. Among so many factors causing RA, cytokines are one of critical to the occurrence and the development of RA.Lugua Polypeptide Injection is abstracted from os cervi and muskmelon seed by contemporary technology, which contain bone induction polypeptide species biological factor, extract matter from muskmelon seed, many kinds of amino acid, organic Calcnd and Phosphorus, and so on. In the experiment, 70 male Wistar rats were divided into 7 groups at radom.10 rats in each. Except normal control group, others Wistar rats were injected about 0.05ml at the right hind paw with Freun's Complete Adjuvant, successfully induced AA animal model. After 7 days, the treated groups were separatately fed with Lugua Polypeptide Injection (4mg/kg,ip), different dosage of Lugua Polypeptide Injection(ig), including 5mg/kg,10mg/kg, 20mg/kg and Aspirin (200mg/kg,ig)every day at the same time for 12 days. During this period, the changs of general status and the swelling of joint have been recorded on different time. After 12 days of treatment, the rats were put to death and their serums were collected. And then observed the adjusting effect of Lugua Polypeptide Injection on IL-2 and TNF-αin serum. At the same time, measured the levels of MDA and SOD in serum, estimated PGE2 in joint organization. Furthermore, the whole blood viscosity, ESR , the White blood count and the peritoneal macrophage phagocytose function were detected respectively. In order to know whether Lugua Polypeptide Injection can lighten the swelling of joint and restrict the damage from inflammation CK and inflammation medium by ig at acute period in AA rats or not.When in RA, ESR has became quick, the White blood count and the content of PGE2 in joint organization has been raised, the content of TNF-αin serum has increased. on the contrary, the content of IL-2 in serum has decreased. Therefor, these phenomenons show that the period of the disease was acute period in RA, and the whole body immunity became decreased due to the content of IL-2 decreased. The result of experiment demonstrate that small and middle dosage of Lugua Polypeptide Injection manifest decrease the swelling of joint in RA, reduce the content of PGE2,TNF-αand increase the content of inflammation CK, stimulating the content of IL-2, getting rid of immune inhibition to adjust the whole body immunity.When in RA, free radical and peroxidized-lipid were made both the structure of joint and the function of joint damaging,because of the hyaluronic acid of mucoitin depolymerize in synovial membrane and cartilage at joint, synovial membrane viscosity change, collagen denature, chondrogenesis degrdn, and so on. Therefore, Peroxidized-lipid increase and/or SOD decrease can accelerate RA. So preventing Peroxidized-lipid is beneficial to anti-RA. The result demonstrated that Lugua Polypeptide Injection can decrease the content of MDA in serum, obviously increase the activity of SOD. These point out that Lugua Polypeptide Injection with intragastric administration maybe through increaseing the activity of free radical eliminating Enzyme, preventing Peroxidized-lipid to prevent RA.The change of blood rheology in RA could caussed the increasing of blood viscosity,consequently induced the form of thrombus and caused many system ruin out of joint. To prevent blood disc gather and blood viscosity are meaningful to prevent RA. We observed that small and middle dosage of Lugua Polypeptide Injection with intragastric administration can markedly decrease the blood viscosity of whloe blood, reduce the risk of many system out of joint in RA.To sum up, Lugua Polypeptide Injection with intragastric administration can increase the activity of SOD and the content of IL-2. At the same time, markedly decrease the content of PGE2, TNF-α, MDA and the peritoneal macrophage phagocytose function, reduce the whole blood viscosity, lighten the swelling of joint in acute period and avoid the damage of many systetem out of joint in RA. It maybe a perspective drug of anti-RA in the future.
