Objective To investigate the influence and mechanism of VEGF and L-arginine on the survival of ischemia skin flap of rat.Methods Forty wistar rats were employed as experimental animal and were established the animal model of random and pedicle skin flap.1. Experimental animal were divided into four groups at random with 10 rats in each group. The four groups are VEGF, L-arginine, VEGF plus L-arginine and control group.2.The colon texture , temperature and capillary recirculate of skin flaps were investigated each day after operation.3.On day 7, the models of flaps were drawn out by transparent graph paper, and the zones of necrosis were overlaid into black. Survival areas of the skin flaps were calculated by computer image analysis software system.4. At 7 days post-operatively, Skin flaps were harvested and fixed with 10% formalin, then embedded in paraddin and stained with HE. Variance of histology, neutrophil counts , subdemic vascular number were evaluated under the microscope.5. The evidence of NO and VEGF expression was detected by immunohistological method.
