Objective:To prepare folate conjugated bovine serum album nanoparticles and observe the targeting effect of folate-BSANP to A549 cells, the cells of human lung adenocarcinoma, on the basis of that folate receptors are frequently overexpressed on some kind of tumor cellular membrane, but they are expressed on normal tissue at a low lever or not. Methods:1. The folate activated by l-ethyl-3-(3-dimethylaminopropyl) cabodiimide muriate was reacted with BSA to get folate-BSA. The unreacted folate was removed by gel filtration. After lyophilyzation, the conjugation rate of folate and BSA was examined. Then it was reacted with calcein to gain F-BSANP. F-BSANPs were observed and took photos under the electron microscope. The diameter of the nanoparticles was calculated.2. BSA was reacted with calcein to prepare BSA nanoparticles (BSANP)3. A549 cells were preincubated in culture dishes, and then were dived into group A, B and C. BSANP, Folate-BSANP and Folate-BSANP plus folate were added in each group, respectively. After 3h, the cells were observed under a fluorescence microscope and took photos.Results:1. The conjugating rate of folate on BSA was 16.05%.2.
