Objective To explore the anti-inflammatory effect of electro acupuncture (EA) and its intervention on COX-2 in anti-inflammation, we observed the therapeutic effect of EA on carrageenan-induced acute inflammation in rats,as well as cyclooxygenase -2 (COX-2) expressions in inflammatory tissues. To explore the connection between regulation of proinflammatory cytokines and inhibition of COX-2 expression by EA,we observed the intervention of EA on interleuk-1 β (IL-1 β ) and tumor necrosis factor-a(TNF- α) concentration and mRNA expression on carrageenan-induced acute inflammation ,and the intervention of EA on COX-2 expression in air-pouch experiment induced by hr(human recombination) IL-1 β and TNF- α .The study will provide a scientific basis for the anti- inflammatory action of EA.Methods The right hind paw of each wistar rat was injected subcuta-neously with 0.1ml of 1% carrageenan to induce acute inflammation model, and the rats were divided randomly into control group and four treatment groups including longterm EA group, immediate EA(EA2) group, indomethacin group and rofecoxib group. Paw edema was measured at 1h intervals. Concentrations of PGE2 in local inflammatory tissues were measured by ELISA. COX-2 mRNA and protein expressions in inflammatory tissues were detected by RT-PCR and Western Blot . Concentrations and mRNA expression of IL-1 β and TNF-α...
