ObjectiveTo investigate the method of establishing subcutaneous chronic inflammation-melanoma model in mice. By observing melanoma’s volume,weight and microvessel density(MVD) to clarify effect of chronic inflammation on melanoma growth and transplant. Lay the foundation for further study about the relationship between inflammatory factor and tumor.Contents1. The establishing of subcutaneous chronic inflammation mice model. The chronic inflammation was induced by injecting two kinds of proinflammatory substances (Lipopolysaccharide or Bacillus Calmette-Gu) into subcutaneous air pouches of mice.In control mice, saline was injected.The progress of inflammation were recorded.2. The establishing of subcutaneous melanoma mice model. After the mouse were disinfected by75%alcohol, suspensions of melanoma cells were inject into the subcutaneous of strech marks.3. The establishing of subcutaneous chronic inflammation-melanoma mice model. After3d the proinflammatory substances was injected into the air pouch or around of glass beads buried under skin the first time, the strech marks of mouse were injected with melanoma cells, and the proinflammatory substances inject every week.Then we compare four kinds of model, chose the best one for the future study.4. The effect of chronic inflammation on planting,growth and migrate. Compare the volume of tumors, infiltration to lung, liver and spleen.5. The effect of chronic inflammation on growth of endothelium-dependent Vascular.After tumor tissue were stained routinely with hematoxilyn and eosin, then analyse MVD of each group.Methods1. The chronic inflammation was induced by injecting two kinds of proinflammatory substances (Lipopolysaccharide or Bacillus Calmette-Gu) into subcutaneous air pouches of mice.In control mice, nomal saline was injected.The progress of inflammation were recorded. 2. We established four kind of models, which contained group group A, group B, group C, group D and group E, according to the difference proinflammatory substances and operation treated to the mouse.3. Tumor’s volume was calculated accuording literature.Tumor weighted by electronic balance.The tissue were stained routinely with hematoxilyn and eosin.hematoxylin.MVD were examined by Hematoxylin-Eosin stain and counted by Weidner method.Results1. The subcutaneous chronic inflammation model could be established by injecting of LPS or BCG into the air pouch.The model can simulate the real situation of chronic inflammation induced by single proinflammation substances.It take about7days for the acute inflammation transform into chronic inflammation.2. We don’t found metastasis in the lung,liver,spleen of the mouse in both experiment group and control group.The difference of the average tumor volume and weight: tumor weight between the group A and control group was statistically different on the21st day. Tumor weight between the group B and control group was statistically different on the15th d.21st day. Tumor volume was on the21d. Tumor weight and volume between the group C,D and control group was statistically different on the9th,21st day.3. The difference of the average tumor MVD. Between group A and control group was statistically different on the9th,15th,21st,27th day.Between group B and control group was statistically different on the9th,15th.21st.27th.day between group C and control group was statistically different on the21st day. Between group D and control group was statistically different on15d.21d.Conclusion1. The model can successfully simulate the real situation of chronic inflammation induced by single proinflammation substances.2. All the four kinds of animal model have same influence on tumor.but they have different proinflammatory substances and operation on theinflammatory position,management. A combination of factors the air pouch freund’adjuvant group was the best.3. Among this four kinds of animal model.we observed chronic inflammation can promote tumor volume and weight growth. 4. Among this four kinds of animal model,we observed that mediators of inflammation can have effect on endothelium-depent vascular of ectopia tumor.
