Objective Cancer is a cell cycle disease,uncontrolled cell proliferation is the basic feature of cancer.Tumorigenesis and its development are closely related to the misregulation of molecular mechanism of cell cycle. CyclinE and CyclinA1 both are important members of a growing superfamily of cyclins ,which,in conjunction with CDK respectively,regulate essential process at G1-S-phase of cell cycle;P27,an essential cell cycle inhibitor of cyclin-dependent kinase,inhibits the activity of CDKs by binding both to cyclins/CDKs and blocks the cell cycle during G1 and S phase; Skp2(S phase kinase -interacting protein 2) is the specific recognition factor for P27 ubiquitination,and ubiquitin-mediated proteolysis of P27 is an important mechanism for activation of cyclinE/CDK2 and facilitates G1/S transition. The study was conduct to analyze the expression levels of cyclinE, cyclin A1 ,P27 and Skp2 in acute leukemia patients bone marrow and combined the clinical data analysis to investigate the roles of these cell cycle regulators in the development and progression in leukemia.The research was expected to provide the basis of identification for chemical therapy.Methods A study of 60 cases and 10 controls was carried out with immunohischemistry method .The expression levels of cyclinE, cyclinA1, P27 and Skp2 in acute leukemia patients bone marrow were examined and combined the clinical data analysis to investigate the abnormal expression of these cell cycle regulators in the development and progression in leukemia. Independent prognosis factors of these cell cycle regulators toward the CR of AL were analyzed by Logistic regression.Result The expression levels of cyclinE, cyclinAl and Skp2 in the de novo and the relapsed group were significantly higher than that of the complete remission and the control group(p<0.05),and the expression levels of cyclinE in the relapsed group...
