Osteosarcoma is the most common primary malignancy of bone, which occurs mostly in juniors and threatens the patients' lives because of its early hematogenous metastasis and then a low 5-year survival rate. So far there is no clear understanding on the etiology and pathogenesis of this neoplasm. To study on the cell cycle regulation and molecular mechanism would benefit to elucidate the tumorigenesis of osteosarcoma and establish a new method for treatment. P27kipl is one of recently discovered and important CKIS (cyclin-dependent kinsse inhibitors)molecules.It negatively regulates the cell cycle through inhibiting CDK (cyclin-dependent kinase).CyclinE is one of positive regulating factors in Gl period. It combines CDK and promotes the DNA synthesis to realize the switch from Gl to S period. Reproduction rate of tumor cell is one of the important parameters which evaluate the biological behavior of tumors. Ki-67 is a better marker for researching the state of cell proliferation. So far, there are few reports on the co-expressions of P27kipl, cyclinE and ki-67 gene in human osteosarcoma. Thirty specimens of human osteosarcoma tissues (OST) were obstained. They have been classified as three grades (I grade 7 cases, II grade 14 cases, III grade 9 cases) according to the criterion of LiuChangMao. The control were 10 cases of osteochondroma tissue (OCT).Immunohistochemical S-P method was used to investigate the biological significance and relationship ofthese genes in OST and OCT. The result are follows:1. expression of P27 protein (1) the positive immunostaining rate in OCT was 80%(8/10), in which weak, moderate and strong positive staining was 2, 3, 3, respectively; the positive immunostaining rate in OST was 33.33%(10/30), in which weak, moderate and strong positive staining were 7, 2, 1, respectively. Comparing the staining rate and intensity of OCT and OST, respectively, there were all significant differences (PO.05) . (2) the positive immunostaining rate of grade I group was 71.43% (5/7), in which weak, moderate and strong positive staining were 3, 1, 1, respectively; the positive immunostaining rate of grade II+III group was 21.74% (5/23), in which weak and moderate positive staining were 4, 1, respectively. Comparing the staining rate and intensity of grade I and II+III grade, respectively, there were all significant differences (PO.05) . (3) the positive and negative immunostaining cases in osteoblast cell type, chondroblast cell type, fibroblast cell type and mixed type were 5 and 9, 2 and 3, 2 and 6, 1 and 2,respectively. Comparing the staining results of the 4 types, there all were not significant differences (P>0.05).2. expression of cyclinE protein (1) the positive immunostaining rate in OCT was 30%(3/10), in which weak, moderate positive staining was 2, 1, respectively; the positive immunostaining rate in OST was 93.33%(28/30), in which weak, moderate and strong positive staining were 6,16,6, respectively. Comparing the staining rate and intensity of OCT and OST, respectively, there were all significant differences (PO.05). (2) the positive immunostaining rate of grade I group was 85.71% (6/7), in which weak, moderate and strong positive staining were 4,1,1, respectively; the positive immunostaining rate of grade II+III group was 95.65% (22/23), in which weak , moderate and strong positive staining were 2, 15, 5, respectively. Comparing the staining rate of grade I and II+III grade, respectively, there was no significant difference (P>0.05); Comparing the staining intensity grade 1 and grade 2+3,respectively,there was significant difference (PO.05). . (3) the positive and negative immunostaining cases in osteoblast cell type, chondroblast cell type,fibroblast cell type and mixed type were 13 and 1, 4 and 1, 8 and 0, 3 and 0, respectively.
