Background:With an increasing incidence, peripheral neuropathy is one of the most common complications of diabetes.It is related to the function loss of sensory,motor, autonomic nerve exhibiting numbness, soreness, dyscinesia,muscular wasting even paralyzed and autonomic nerve functional disturbance. It's not clear about the pathogenesis and not find the effective treatment and drugs yet.With more experimentive research about diabetes mellitus, although it remains elusive about the pathogenesis of DN,several mechanisms may play an important role leading to the disturbance and have an interaction: vascular, metabolism,nerve growth factor, autoimmune and inflammation. It contributes to seek therapies and effective drugs for diabetic neuropathy. As a member of the neurotrophic family, The best studied is nerve growth factor.It has been found that there is a reduction in the availability of NGF in diabetes and it can amend the disturbance of the nerve to supplement the NGF.Substance P is a neuropeptide regulated by the NGF.It has been implicated in diverse and widespread activities, including vasodilatation and nociception, all of which are perturbed in diabetic neuropathy. There is increasing evidence that there is a deficiency of NGF in diabetes, as well as the dependent neuropeptides substance P (SP). It seems that and inistration of NGF may be of value in treating the early diabetic Neuropathy in animals,but not a ideal unitary clinical treatment. There are many good results of the research on DN with TCM refers to various links and levels.Objective: To study the effect of Chinese medicine"QiTengTong LuoYinDecoction"on diabetic neuropathy(DN), establish the foundation ofclinical application and provide new research clues of prevention on DN with Chinese medicine.Method:Models of diabetic rats are induced by single intra- peritoneal injection of Streptozotocin(STZ)and fed with"Qiteng tongluoyin".Weight,fasting blood glucose,glycohemoglobin (HbA1c) of DN 4 wk or 8 wk rats were observed.The morphology of pancreatic gland and sciatic nerve were investigated. The expression of endogenous nerve growth factor (NGF)and substance P(SP)in sciatic nerves were observed by immuno- histochemistry.Results:The rats of the model group are weaker than the rats of the treatment group,and in a higher blood glucose level and glycohemoglobin. The morphology of the sciatic nerve and pancreatic gland of the rats of treatment group was lighter compared with the model group.Expression of NGF and SP in sciatic nerve of treatment group rats are more than Diabetic Model(DN) group(P<0.05).Conclusions:"Qitengtongluoyin"has a preventive and treatment effects on pathological changes of sciatic nerve of DN rats.It's pissible mechanism may be associated with the promotion of expression of endogenous NGF and SP.
