Purposes: To study the expression of apoptosis related gene survivin and its relation with expression of bcl-2 , p53 protein in epithelial ovarian cancer.Methods:Using SP immunohistochemical technique, we examined the expression of survivin, Bcl-2 and p53 protein in 35 cases of epithelial ovarian cancer,10 cases of benign tumor and 10 cases of normal ovary tissue.The correlation between the expression of survivin protein and clinicopathologicai characteristics in epithelial ovarian cancer was analyzed.Results: (1)survivin positive expression rate in epithelial ovarian cancer was 62.86% (21/35) ,p53 positive expression rate was34.29% (12/35), Bcl-2 positive expression rate was 57.14%(20/35) ;the positive expression rate of survivin, p53, Bcl-2 in epithelial ovarian carcinoma tissues was significant higher than in benign tumor tissues and normal ovary tissues. (2) survivin expression displayed a positive correlation with p53(P<0.05).(3)survivin expression displayed a positive correlation with Bcl-2 (P<0.05).(4)Overexpression of survivin was related to the tumor grade and clinical stage. There was no relationship between surviving protein expression and pathological type and lymphatic metastasis. The expression of Survivin was correlated with unfavorable outcomes. Survivin positive cases were associated with Bcl-2 and p53 expression.Conclusion: (1)Survivin is an inhibitor of apoptosis that is highly expressed in epithelial ovarian cancers (EOC),but lowly or not in benign ovarian tumor and normal ovarian tissue . Survivin gene may play an important role in pathogenesis of ovarian cancer.(2)Apoptosis related genebcl-2 may have a synergic role and p53 have an antagonistic role with survivin gene in formation and progression of ovarian cancer.(3) The combinating detection of survivin, p53, bcl-2 expression may assess the clinical prognosis of epithelial ovarian carcinoma.Purposes: To investigate the relationship of the expression of survivin and p53 in epithelial ovarian cancer cells with chemosensitivity.Methods: Surgical samples of epithelial ovarian cancer, diagnosed by pathology,originated from The PLA General Hospital. The cancer sample cells were cultured separately in the incubator at 370C,5%C02 in vitro. The rate of inhibition of cancer cells by 4 kinds of anticancer drugs, which were FTX, TXT, DDP and ADM were assayed by MTT method. Immunocytochemistry was used to detect the expression of survivin and p53 in the cancer cells.Results: Sensitivity rate of 12 epithelial ovarian cancer cells in 0.1 X PPC within 72h in vitro were 66.7%(PTX), 41.67%(TXT), 58.3%(DDP) and 33.3%(ADM) The rate of inhibition of PTX ,TXT,ADM and DDP were significantly higher in the survivin negative cancer cells than in the survivin positive cancer cells . The chemosensitivity of survivin (-)p53(+)for PTX, TXT is higher than survivin (+)p53(-) and survivin (+)p53(+), ADM is higher than survivin (+)p53(-)and survivin (-)p53(-);surviving (+) p53(-) for DDP is higher than survivin(+)p53(+)and survivin (-)p53(+).Conclusions: The expression of surivivn is related with the chemosensitivity of the epithelial ovarian cancer cells, and it may become one of the markers for judging the effect of chemotherapy in clinic.The combinating detection of surivivn expression, p53 may be taken as an index of the selection of anticancer drugs.
