Objective: To compare the efficacy of heart preservation of HTK solution,FWM solution and St.Thomas-II solution following 4,6 and 8 hours hypothermia preservation in the isolated rat heart mode.Methods: Male SD rats (n=54) were divided randomly to three groups: HTK, FWM and ST-II(n=18 in every group), and they were divided again to three groups(n=6) according to difference of storage period. The heart were harvested and mounted on the Langendorff apparatus to perfuse with K-H buffer solution to estimate baseline hemodynamic values (HR,LVSP, + dp/dtmax, -dp/dtmax, CF) and the leakage of enzymes of the myocardial tissue (CK-MB,LDH) .Then they were arrested and stored in each preservation solution (4°C) for 4hours,6 hours or 8 hours. At the end of the storage period, the rat hearts were resumed and reperfused in Langendoff model. Which the recovery rate of the hemodynamic values,the water content and the leakage of enzymes of the myocardial tissue were measured. At the end of the reperfusion, the myocardial specimen were made to be observed the changes of structural through light microscope.Results: (1) In the same storage period, the hearts stored in HTK or FWM have a higher recovery rate of the hemodynamic values and a lower level of water content and the leakage of enzymes of the myocardial tissue than those stored in ST- II (P < 0.05). And the changes of myocardial structural in ST- II group were more remarkably than in HTK or FWM group. There was no difference of all index existed between HTK and FWM group stored 4 or 6 hour (P > 0.05), but there was significant difference stored 8 hour(P < 0.05).(2) The heart after 8 hour cardioplegia in HTK group have a lower recovery rate of the hemodynamic values, a higher level of water content and a more leakage of enzymes of the myocardial tissue than those stored 4 or 6 hour P < 0.05).Conclusion: The results suggest that HTK solution and FWM is more effective than St.Thomas- II solution for hypothermic myocardial preservation in the same storage period, and HTK solution is the most effective one. And the efficacy of myocardial preservation of HTK solution changed with prolonged storage period.
