Objective To analyse the result of the TGF- β1 effect in the fibroblastdifferentiated from bone marrow-derived mesenchymal stem cells by morphology and immunohistochemistry. and compare the difference between normal human and CMPD Patient's bone marrow fibroblasts from bone marrow-derived mesenchymal stem cells. Methods bone marrow MSCs were separated from one normal human(IDA patient) and one CMPD patient by density-gradient centrifugalization. Cultured with medium consisted DMEMof lesser glucose and 10% fetal calf serum(FCS). Incubated at 37°C, 5% CO2 condition. Digested by 0.25% trypsin and 0.02% EDTA, differentiated by the TGF-β1, identified by flow cytometry and morphology, evaluate the expression of collagen fibrils by modified Masson trichrome staining, assessthe cell matrix potein by immunohistochemistry. Results Combined with density-gradient centrifugalization and adherence, high purity and homogenicity MSCs can be obtained. 5ng/ml TGF-β1 promote the expression of collagen and vimentin,. Conclusion MSCs present with fusiform, triangulum, polygon shape, express CD29,CD44,CD166, not CD14,CD34,HLA-DR, CD45. TGF- β1 promote fibroblast differentiate from MSCs.
