OBJECTIVE To explore the expression and significance of monocyte chemotact-ic protein-1(MCP-1) , Cyclooxygenase-2 (COX-2) in mayometrinl during labor and placental and their relationship with the onset of labor, cervical ripening and dilation.METHOD Biopsy specimens were taken from the lower uterine segment of 62 women undergoing cesarean section at term who were divided into the group of cesarean section with (42 cases) and without (20 cases) uterine contraction. The stage of cervical dilatation 30 minutes before the time of cesarean section of uterine contraction group was < 2 cm ( group A, n = 10), 2 to< 4 cm ( group B, n = 10), 4 to6 cm ( group C, n = 10) or > 6 cm ( group D, n = 12). The Real-time Fluorescence Quantitative PCR (FQ-PCR) method was used to detect the expression of the target gene MCP-1,COX-2 and the internal standard gene GAPD gene. The Ct (cycle threshold) datas were read from the software on the LightCycler2.0. If the difference of the average Ct had the statistical significance, then the 2-△△Ct formula was used to accomplish the relative quantification.RESULTS In the group of cesarean section with and without uterine contraction, MCP-1 and COX-2 tended to increase with the dilatation of cervix. There was statistical significancebetween them by the t test (P<0.01). The expression of MCP-1 and COX-2 mRNA were significantly greater at group C and group D than group A and group B (P<0. 01). There were statistical significance (P<0.05) between group A and group B by the q test, and there was statistical significance (P<0.05) between group C and group B of the expression of COX-2 mRNA, but no statistical significance (P>0.05) between group C and group D of the expression of MCP-1 mRNA, which mean no significant distinction of MCP-1 levels between group C and group D. The expressions of MCP-1 and COX-2 were correlated with cervical dilatation.CONCLUSION To detect the expression of MCP-1 and COX-2 mRNA during the different stage of cervical dilation in the lower uterine segment and placenta tissues with and without uterine contraction,the result displayed that MCP-1 and COX-2 expressed in all the puerperants, MCP-1 and COX-2 tended to increase with the dilatation of cervix. Cervical dilatation was associated with up-regulation of MCP-1 and COX-2 in the lower uterine segment and placenta tissues,which prompted that there were close correlation between MCP-1,COX-2 and labor,and played an important role during the delivery. MCP-1 and COX-2 had positive correlation, which hint that they played synergistic effect during the stage of cervical dilation and had co-effection in the process of delivery.To approach the expression and significance of MCP-1 and COX-2 in the delivery will be some instructions in the cervical ripening and dilation.
