OBJECT: The purpose of the study was to explose the relationship between autonomic dysreflex (AD) and the expression of calcitonin gene-related peptide (CGRP) at spinal dorsal horn in rats after spinal cord injury (SCI) 0~1 5 days. To research the intervention of CGRP antagonist CGRP (8-37) in AD. To explorer the treatment for AD after SCI.METHODS : (1)Made SCI animal model with Wistar rats by spinal cord transaction at thoracic 4 (T4). Control group was just exposed lamina. Observed the blood pressure change value when clipped skin superior transactiotn plane, inferior transaction plane, rat tail, and distended colon with 0.3ml balloon, and changed posture after SCI 0, 3, 6, 9, 12 andl5 day. Then we investigate the expression of CGRP at spinal cord dorsal horn (T5~T8,T9~T12,T13~L2).(2) Both experimental group and control were made SCI animal model by spinal cord transaction at thoracic 4 (T4). The experimental group was given CGRP (8-37) via subarachnoid cavity at the 13th days after SCI ,while the control group received saline. Research the blood pressure change value when rats were clipped skin inferior transaction plane ,rat tail, and distended colon with 0.3ml balloon. Then investigate the expression of CGRP at 14th day.RESULTS : (1)The blood pressure change value after every stimulus, except the rats were clipped skin superior transaction plane, had relationship with time after SCI in the experimental group (P<0.05). There were not relationship between the expression of CGRP of experimental group and that of control group(P>0.05). There were relationship among the expression of CGRP in every section in control group(P<0.05). There were relationship among the expression of CGRP in every section in control group (P<0.05).The expression of CGRP in experimental group changed with time after SCI along quadratic function.There were relationship between the expression of CGRP and the blood pressure change in the experimental group , when the rats were clipped skin inferior transaction plane ,rat tail, distended colon with 0.3ml balloon and changed posture(P>0.05).(2) blood pressure change value was compared between the control group and experimental group with statistically significant difference.CONCLUSION : (1) AD gradually aggravated in the first 15 days after SCI, and is closely related with the expression of CGRP. High expression of the CGRP AD is an important factor of AD, and changed with time after SCI along quadratic function.(2) The experiment showed that CGRP (8-37) could reduce AD and as a new treatment way for AD.
