Experimental Study Of Effect On Hepatic Tissue MMP-1 And TIMP-1 Of Curcumin On Hepatic Fibrosis Tissues In Rats

Objective:To observe the effect of Curcumin on hepatic tissue matrix metalloproteinase-1(MMP-1) and tissue inhibitor of matrix metalloproteinase-1(TIMP-1) of hepatic fibrosis tissues in rats and investigate the relationship between different dosage and curative effect to further discuss the mechanism of antifibrotic action and the appropriate dosage.Methods:models of hepatic fibrosis were established by hypodermic injection of tetrachloride and filling the stomach of ethyl alcohol. Curcumin of 10mg,20mg,40mg per 100 gram weight of rat were given to these rats respectively at the same time.Normal,fibrotic and treated groups were made as controls.After 20 weeks,all the rats were executed and the blood were kept for detecting HA,LN,PC-â…¢,â…£C. Livers were extirpated for pathologic examination including HE,Van-Gieson and Gomori stains.The content of hepatic tissue MMP-1andTIMP-1 were detected by immunohistochemical technique.Result:Serum levels of HA,LN,PC-â…¢,â…£C hepatic tissue TIMP-1 of fibrotic group increased significantly compared with normal group,which were depressed obviously in large and center Curcumin groups(P<0.01) . HA,LN were depressed obviously in small Curcumin group (P<0.05) . Staining of HE,Van-Gieson and Gomori showed that degree of hepatic fibrosis in large and center Curcumin groups were improved obviously in fibrotic group. The content of hepatic tissue MMP-1 was increased by large and center Curcumin groups(P<0.01).Conclusions:1.Curcumin can increase MMP-1 of hepatic fibrosis tissues in rats, reduce deposition of hepatic tissue TIMP-1,degree extracellular matrix(ECM) of hepatic fibrosis.2.The Curcumin anti-hepatic fibrosis existence effective dosage scope, in the certain scope,the anti-hepatic fibrosis function and the dosage doesn't present the relevance.3.The Curcumin has the function of treatment hepatic fibrosis.
