Objective:To observe the neuroprotection of PROG on rat retinal ganglion cells in acute ocular hypertention model. Methods: 35 femal Wistar rats without eye diseases were divided into 3 groups: group I:normal group(5rats,10eyes) ,other 30 rats was induced in left eye by forcing perfusion of balanced salt solution into the anterior chamber as acute ocular hypertention model.Right eyes were self-control eyes . 30 rats were divided into group II:PROG treated group (15 rats), group III:balanced salt solution treated group(15 rats).1 hour before we setting up the model of acute ocular hypertention and 12 hours,24 hours,48hours after we setting up the model, rats had intraperitoneal infections of PROG. (2.0mg/kg, 1.0ml) in group II respectively. Rats had intraperitoneal infection of balanced salt solution in group III at the same time. 48 hours later , Rats were executed . Not standard eyeballs were discard and 10 eyeballs were selected in each group.The eyeballs were enucleated and fixed in 4% paraformaldehyde .The retinal ganglion cells were detected by HE staining and Thy-1.1 immunohistological staining. The apoptosis cells were detected by TdT-dUDP terminal nick end-labeling (TUNEL)technique.Result: The number of RGCs of model eyes in group II and III was less than that of group I and self-control eyes,there was significant diferences (P<0.05). But the number of RGCs of model eyes in group II was more than that of group III,there was significant diferences (P<0.05). The number of TUNEL-positive cells of model eyes in group II and III were more than that of group I and self-control eyes,there were significant diferences (P<0.05). But the number of TUNEL-positive cells of model eyes in group II were less than that of group III,there was significant diferences (P<0.05). Conclusion:PROG could provide neuroprotection for RGCs in rat acute glaucoma model.
