Objective To investigate the relation of the tumor necrosis factor beta (TNF-β) gene polymorphism to HBV-induced liver fibrosis in china GanSu province.Methods A case control research was done on 146 HBV-induced liver fibrosis patients and 138 healthy individuals as control group. DNA was extracted from peripheral blood leukocytes by DNA extraction kit. TNF-βgene polymorphisms were studied by using nested PCR and allele specific PCR(AS-PCR) technique ,in order to knowing distribution of TNF-βgene polymorphism of GanSu province. Simultaneously fibrotic markers were detected by Farr's technique, comparing its and genotype by one-way analysis of variance.Results 1. HBV-induced liver fibrosis group, the frequencies of the TNF-βCC,CA, AA genotype respectively is 24.0%,47.9%,28.1%; control group is 37.0%,45.7%,17.4%.AA genotype increased significantly in HBV-induced liver fibrosis patients compared with healthy controls having statistics significance (x~2 =7.572,P < 0.05). The frequencies of the allele C and A in patients group respectively is 47.9% and 52.1%,in control group respectively is 59.8% and 40.2%;A allele increased significantly in HBV-induced liver fibrosis patients compared with healthy controls having statistics significance (x~2=7.996,P<0.05). 2. Besides AA genotype IV.C, the fibrotic markers of the different TNF-βgenotype does not have statistics significance (P > 0.05).Conclusion TNF-βgene polymorphism was associated with HBV-induced liver fibrosis. The allele A might confer risk for the development of HBV-induced liver fibrosis.
