Objective To investigate the expression of Runx3 and p21 protein in gastric carcinoma and cancer-adjacent tissue. Explore their action and relationship between them in the cause of growth, development, infiltration and metastasis of gastric carcinoma so that useful marker can be obtained for diagnosis of pathology, treatment and prognosis of gastric carcinoma clinically.Methods The specimens of 50 gastric carcinoma, 22 heteromorphism hyperplastic tissue and 20 normal gastric mucosa were studied with conventional HE and immunohistochemistry staining(SABC) by which Runx3 and p21 were detected.Results The positive expression rates of Runx3 and p21 protein were 95% (19/20),59.1%(13/22),34% (17/50) and 85% (17/20),54.5% (12/22),42% (21/50) in gastric carcinoma, heteromorphism hyperplastic tissue and normal gastric mucosa tissue respectively. Significant difference of positive expression rates of Runx3 and p21 were calculated among them statistically (P<0.05). And no significant difference of positive expression rates of p21 (P>0.05) but of Runx3 (P<0.05)were calculated between gastric carcinoma and heteromorphism hyperplastic tissue statistically. The level of Runx3 expression was involved with differentiation grade, lymph node metastasis and distant metastasis. It was higher in high differentiation grade, no lymph node metastasis and no distant metastasis group than in low differentiation grade, lymph node metastasis and distant metastasis group, (P<0.05) . The level of p21 expression was involved with differentiation grade, lymph node metastasis, distant metastasis and invasive depth of gastric carcinoma. It was higher in high differentiation grade, no lymph node metastasis, no distant metastasis and invasive depth(T1+T2)group than in low differentiation grade, lymph node metastasis, distant metastasis and invasive depth(T3+T4)group, (P<0.05). Positive correlation was deduced between Runx3 and p21 in gastric carcinoma, heteromorphism hyperplastic tissue and normal gastric mucosa tissue. And the correlation coefficient r was 0.998, P<0.05.Conclusions 1,Significant difference of positive expression rates of Runx3 was calculated in gastric carcinoma, heteromorphism hyperplastic tissue and normal gastric mucosa tissue statistically (P<0.05). The level of Runx3 expression was involved with differentiation grade, lymph node metastasis and distant metastasis(P<0.05). It is demonstrated that Runx3 protein may play a important role in the growth and development of gastric carcinoma.2,The positive expression rates of p21 was lower in gastric carcinoma and heteromorphism hyperplastic tissue than in normal gastric mucosa tissue statistically (P<0.05). The level of p21 expression was involved with differentiation grade, depth of infiltration, lymph node metastasis and distant metastasis(P<0.05). It is demonstrated that p21 protein may be a earlier incident in the growth of gastric carcinoma.3,Positive correlation of expression between Runx3 and p21 was calculated in gastric carcinoma. The low expression of Runx3 and p21 was one of the important reasons in the growth and progression of gastric carcinoma. Both Runx3 and p21 may play a cooperative role in the growth of gastric carcinoma. Therefore, Runx3 and p21 protein may acts as markers predicting biological behaviors of gastric cancer.
