Objective: It has been clarified that cardiovascular risks were correlated with IGT.Activated platelet played an important role in thrombosis, atherosclerosis and inflammatory reaction .There were few reports on platelet function in the stage of IGT.The aim of the study was to investigate the change of platelet activity in subjects with impaired glucose tolerance and its influencing factors.Methods: 33 subjects with IGT,30 patients with type 2 diabetes mellitus and 30 normal control were enrolled in the study.Percentage and mean fluorescence intensity of CD62P,PAC-1 were measured as indices of platelet activation using whole blood flow cytometry.Changes of count of blood platelet,mean platelet volume(MPV), platelet distribution width(PDW) andplatelet-large cell ratio (P-LCR) were analyzed with blood cell counter.TC,TG,HDL-C,LDL-C,VLDL-C,FPG and FIns were measured simultaneously.Using HOMA-IR to evaluate insulin resistance and HOMA-βassess beta-cell function.Results:1.IGT and Subjects with diabetes had higher levels of CD62P (%) CD62MFI and PAC-1(%) compared with the NC group:CD62P(%)(IGT:4.02±2.35,DM:5.25±4.88,NC:1.24±0.31,P<0.01 respectively),CD62MFI(IGT:20.29±5.37,DM:20.41±2.58,NC:14.68±5.16,P<0.05 respectively) and PAC-1 (%) (IGT:24.99±14.61,DM:25.21±21.46,NC8.67±6.29 P<0.01 res-pectively).CD62P(%),CD62MFI and PAC-1(%)have no statis-tical difference between IGT and DM group.2.CD62P(%),CD62PMI,PAC-1(%) and PAC-1MFI expression on platelet was no obviously change(P>0.05) after oral glucose loading 2 hours than fasting stage in IGT.3.IGT and Subjects with diabetes had higher levels of MPV.PDW and P-LCR compared with the NC group:MPV(IGT:10.31±0.87,DM:10.59±0.93,NC:9.52±0.9,P<0.05 respectively), PDW(IGT:12.62±1.06,DM:12.91±1.87,NC:11.32±1.58,P<0.05respectively)and P-LCR(%)(IGT 0.28±0.065,DM:0.29±0.06,NC:0.22±0.067,P<0.05respectively).MPV,PDW and P-LCR(%) have no statistical difference between IGT and DM group. 4.In IGT group,the percentage of CD62P was positively related to DBP(r=0.367,P=0.036) and BMI(r=0.412,P=0.017);CD62PMFI was positively correlated withMPV(r=0.417,P=0.016PDW(r=0.364,P=0.0038)and P-LCR(r=0.433,P=0.012).The percentage of PAC-1 was positively related to LDL-C(r=0.37,P=0.004), MPV (r=0.316,P=0.014) and P-LCR (r=0.29,P=0.025);PAC-1MFI was positively correlated with MPV(r=0.367,P=0.036) and P-LCR(r=0.389,P=0.025).5.By stepwise regression analysis in IGT group,P-LCR(b=33.891,P=0.011),LDL-C(b= 4.959,P=0.009),TC(b=2.373,P=0.068)and BMI(b=0.225 , P=0.017) were con-tributors to CD62P;P-LCR(b=19.435,P=0.025)MPV(b=5.537,P=p.043)and DBP(b=0.376P=0.093) were contributors to PAC-1.Conclusions:1.The levels of CD62P(%),CD62PMFI and PAC-1(%) expressions were increased in IGT group.There was same degree of platelet activation in subjects with IGT as DM.2.CD62P and PAC-1 expressions on platelet was no obviously affected after oral glucose loading 2 hours in IGT group.3.MPV,PDW and P-LCR of platelet were higher in IGT group. MPV,PDW and P-LCR were positively correlated with CD62P and PAC-1 and can reflect platelet activation in IGT.4.LDL-C,TC,DBP and BMI were contributors to CD62P and PAC-1 expressions on platelet in IGT.
