Objective To study the growth-inhibition effect and mechanism of permanent iodine-125 interstitial implants for the treatment of C6 malignant glioma in rats,to supply theoretical evidence for the use of the treatment in clinical,to supply new and available method for the treatment of malignant glioma in clinical.Methods C6 brain malignant glioma cells were cultured in vitro.SD rat intracranial brain malignant glioma models were established using stereotactic methods.Then 24 rats were divided to a treatment group(n=12) and a control group(n=12) with random principle.BT-125-I Model Iodine-125 seed with apparent activity of 0.9mCi was implanted into the tumor in the treatment group;whereas the rats in the control group were not treated.Observing the survival rate of the rats,the tumor growth,HE dying,Bcl-2 and Bax immunohistochemical dying after 14 days in order to expatiate the mechanisms of iodine-125 seed's brachytherapy.Results 2 weeks after the C6 brain malignant glioma cells implantation,tumor was found in the brains of the rats.The weight of tumor in control group and treatment group is 0.49±0.06 and 0.34±0.03 separately when experiment is over,they have significant differences(P< 0.01).Tumor growth can be inhibited obviously by iodine-125 in treatment group inhibited rate of iodine-125 is 30.7%.Immunohistochemistry results show that grey scale of Bcl-2 is 126.60±8.26,93.83±6.17(P<0.01) and separately in treatment group and control group,the expression of Bcl-2 protein can be dramatically decreased by iodine-125.Grey scale of Bax is 78.73±5.35,80.50±6.85(P > 0.05) and no obviously influence is found on Bax expression. So iodine-125 can degrade the rate of Bcl-2/Bax. Conclusions Permanent iodine-125 interstitial implants for the treatment can inhibit the growth of C6 malignant glioma dramatically.The anti-tumor mechanism of iodine-125 may be in association with its down-regulating the expression of Bcl-2 protein and reduction the ratio of Bcl-2/Bax.
