To investigate the anticancer effect of Houttuynia cordata, a traditional Chinese medicine, through apoptosis pathway.The inhibitory effect of Houttuynia cordata on viability and proliferation of HeLa cell were detected by Colorimetric CCK-8 assay. The morphological change of Cell was investigated by light microscope and Hoechst33342. Cell cycle arrest and apoptosis were determined by flow cytometry. The expressions of proteins related to apoptosis were investigated by western blotting. The methanol extract of Houttuynia cordata was sequentially partitioned using organic solvents of different polarities to afford water, n-butanol, 60% methanol, n-hexane, and dichloromethane fractions. The IC50 of the 5 fractions are 393±1.6, 93±1.6, 85±1.7, 170±1.5 and 34±0.4μg/ml. n-butanol, 60% methanol and dichloromethane fractions displayed strong inhibitory effect on proliferation of HeLa cells in time-dependent manner with apoptotic morphological change. Treatment with n-butanol, 60% methanol and dichloromethane fractions lead to an increase of percentage of cells in G2/M phase and Sub-G1 phase. Houttuynia cordata extract induced cytochrome c release from mitochondria into the cytosol and subsequent procaspase-9 activation.Those results suggest for the first time that Houttuynia cordata extract induce the apoptosis and cell cycle arrest throngh mitochondria pathway.
