In our study, the anti-tumor effect of phycobiliprotein from Gracilarialemaneiformis, collected from the sea area around Nan'ao Island, Shantou city,Guangdong province was studied. Throuth the methods of CAM, MTI assay flowcytometry, annexin v-fitc/PI fluorescence staining ,DNA isolation analyses andinfluence of PE on Sarcoma180 mice to discuss the effection and mechanism of theanti-tumor activity of PE.Using tissue- trituration method, Hydroxyapatite and Sephadex G-100 column toextract and purify the Phycobiliprotein, the purity can reach 3.86.Through the method of chicken embryo chorioallantoic membrane cam (CAM) todetect the inhibiting angiogenesis effect of PE. The result showed that PE had aneffectively inhibiting angiogenesis effect and had a dose-response relationship.The viability of cell lines Hela, EC109, and vascular endothelial in vitro wasexamined by MTT assay. The result showed that PE could obviously inhibit thegrowth of Hela cell with a fair dose-effect relationship, the inhibitory rate could reachmore than 70%. But had little inhibitory effect on EC109 cell and had certainpromoting effect on the growth of MVC cell. Those showed that PE had selectivity ondifferent cells. These probably because of the signal channel of different cells,physiology and biology characters of different cells and the different PE absorbableability. The result of flow cytometry showed that PE could block the cell cycle atG2/M phase, decrease its ability of cell division. Inhibit its ability of cellmultiplication. The result of annexin v-fitc/PI fluorescence staining showed that at thedosage between 0~10ug.ml-1, with the increasing of the PE dosage and treating time,what showed that apoptosis cells and later stage apoptosis cells or necrosis cellsincreasing. The result of DNA Ladder showed that after treating 24hours by PE, therewas light "smear" electrophoresis between 200~400bp.The anti-tumor experiments indicated that the phycobiliprotein could significantlyinhibit the growth of Sarcoma-180 in mice. At the dose of 300mg.kg-1 when givenirrigatively (ig), the inhibition rate was 49.29%. The tests of superoxide dismutase(SOD) activity in red blood cell and lipoperoxide (LPO) level in liver homogenatesuggested that the anticancer effect of phycobiliprotein might be related to itsantioxidation.
