Objective: To explore the relationship between extron of hURAT1 gene polymorphismsand familial gout in the littoral extent of Shandong Han people.Methods: Polymerase chain reaction method was used to amplification thesequence. Then the product of PCR was cleansin,sequencing in 61 patients with primary gout and91 healthy persons in familial gout and 100 healthy controls. The difference of their frequencieswas analyzed by Chi-square test.Results: (1) There were single nucleotide polymorphism on the first and second andeight exon in the object of research. (2) The statistic results of genotype and allele in everysite:①There was no statistic difference in the frequencies of genotype and the frequencies ofallele in-948,-1093,-2524,-2449,-8438,-9555,-9681,-10247 site between A and B andC groups(P>0.05).②There was no statistic difference in the frequencies of genotype in-2093,-2442 site between A and B and C groups(P>0.05) ; There was no statistic difference in thefrequencies of allele in 2093 site between A and B groups(P>0.05), The frequencies of allele Tin Agroup were significantly higher than those in C group. (OR=2.06, P=0.048); There was nostatistic difference in the frequencies of allele in 2442 site between A and B groups(P>0.05),The frequencies of allele G in A group were significantly higher than those in C group.(OR=1.939, P=0.01).③The frequencies of genotype of 1105 site in Agroup were significantlyhigher than those in C group (OR=2.533,P<0.017), The frequencies of allele T in A group weresignificantly higher than those in C group. (OR=2.669, P=0.003); The frequencies of genotype of2448 site in A group were significantly higher than those in C group (OR=4.385, P=0.000), Thefrequencies of allele C in A group were significantly higher than those in C group. (OR=1.939,P=0.01).Conclusion: There were single nucleotide polymorphism on the first and second andeight exon in the object of research. Both the site of 1105 and 2448 in hURAT1 gene maycontribute to the development of primary gout in the littoral extent of Shandong Han population.
