Object: To compare the diagnosis value of lung adenocarcinoma by traditional tumor marker CYFRA21-1, CEA, CA125,NSE with serum protein markers selected by surface-enhanced laser desorption ionization time of flight mass spectrometry.Method: serum samples of 71 lung adenocarcinoma patients and 71 healthy persons with matched gender, age and smoking history were analyzed by SELDI-TOF- MS to select relatively specific serum protein markers and chemo-luminescence to analyze the level of serum tumor makers CYFRA21-1, CEA, CA125, NSE.Sensitivity and specificity were compared with combined detection of these proteins and tumor markers.Results: Five highly expressed protein markers were identified by SELDI-TOF-MS with the relative molecular Weights of 4047.79,4203.99,4959.81,5329.30,7760.12.Among these proteins, the protein with the relative molecular Weight of 4047.79 has the highest sensitivity and specificity (90.14%, 97.18%). And the sensitivity (92.96%) and specificity (90.14%) of combined detection of the five proteins (three items positive) are superior to the combined detection of CYFRA21-1, CEA, CA125, and NSE.Conclusion: SEIDI-TOF-MS techno1ogy is a quick,easy,convenient,and high-throughout analyzing method capable of selecting relatively specific, potential tumor biomarkers from the serum of lung adenocarcinoma patients with superior value over traditional tumor marker in the screening and early diagnosis of lung adenocarcinoma. Though this technology is not consummate, it may have attractive prospect with the potential of clinical application.
