Objective:①To analyze the differential expression of serum proteins mass spectra between healthy controls and nasopharyngeal carcinoma(NPC) patients using Surface enhanced laser desorption ionization time of flight mass spectrometry(SELDI-TOF-MS)and estabish a diagnosis model for NPC. To analyze the differential expression of serum proteins mass spectra among different clinical features.②To analyze the serum proteins mass spectra before treatment and estabish prediction model for the response of radiochemotherapy. To analyze the changes after radiochemotherapy.③To analyze the differential expression of serum proteins mass spectra between healthy controls,intial diagnosis,complete remission and relapse NPC patients,aiming at idenitifying serum biomarkers for monitoring NPC relapse.Methods:①SELDI technology and CM10 protein chip were uesd to analyze mass spectra of serum samples from 102 healthy controls and 118 NPC patients,BioMarker Wizard and Biomarker Pattern Software (BPS) were used to analyzed the data and establish diagnosis model. The model was further evaluated by a blind test with another 54 healthy controls and 41 NPC patients. Then this technology was used to analyze the differential expression of serum proteins mass spectra in patients grouped by stages, clinical classification and EB virus infection;②SELDI technology and CM10 protein chip were uesd to analyze mass spectra of pre-treament serum samples from 35 partial remission and 44 complete remission in NPC patients stagingⅢ~Ⅳa. BioMarker Wizard and Biomarker Pattern Software (BPS) were used to analyzed the data and established a prediction model for the response of radiochemotherapy. Later,the same technology was used to analyze the changes of serum proteins mass spectra before and after radiochemotherapy in 30 NPC patients.③SELDI technology and CM10 protein chip were uesd to analyze the differential expression of serum proteins mass spectra between 118 healthy controls,102 intial diagnosis, 11 complete remission and 17 relapse in NPC,and BioMarker Wizard and Biomarker Pattern Software (BPS) were also applided to idenitify serum biomarkers for the monitoring NPC relapse.Results:①68 protein peaks were significantly different between healthy controls and NPC patients(P<0.05), a primary diagnosis model of NPC was set up consisting of 5 protein peaks with M/Z values of 4181Da,4298Da,6450Da,13775Da and 3276Da. Blind test confirmed a sensitivity of 97.56%,specificity of 88.89% and accuracy of 92.63% respectively. M/Z values of 10286Da,7596Da and 8149Da were significantly different between ascending type and descending type of NPC(P<0.05);M/Z value of 9354Da and 4596Da were significantly different between EB negative and EB positive(P<0.05),no significantly different protein peaks were found between different stages of NPC;②No significantly different protein peaks were found in pre-treament serum samples between 35 partial remission and 44 complete remission cases stagingⅢ~Ⅳa after radiochemotherapy. M/Z values of values of 9296Da,7794Da,8175Da,7935Da and 4182Da decreased after radiochemotherapy,close to that of healthy controls, M/Z values of 3205 Da,3276Da and 2949Da were increased after radiochemotherapy,further from that of healthy controls;M/Z values of 13775 Da,4299 Da,6453 Da,6852Da were constantly lower than the that of healthy controls before and after radiochemotherapy.③M/Z values of 7794 Da incrased in intial diagnosis and relapse of NPC,M/Z values of 4182 Da decreased in intial diagnosis and NPC relapse; M/Z values of 4299Da,6453Da and 6852Da were constantly lower in intial diagnosis,complete remission and NPC relapse than healthy controls.Conclusions:①T he results suggest that SELDI-TOF-MS serum protein profiles could differentiate NPC patients from healthy controls with a high sensitivity and specificity and may was promising in clinical application. SELDI could be used to discover some characteristic of serum proteins relates to different clinical features. ②No significantly different protein peaks were found in pre-treament serum samples between partial remission and complete remission patients. Significantly different protein peaks can be found before and after radiochemotherapy;③SELDI results indicated that two protein peaks in serum proteins can monitor the relapse of NPC,some protein peaks were constantly lower in intial diagnosis,complete remission and relapse NPC patients.
