ObjectiveInterleukin-1βcan cause a dose-dependent stimulation on the c-myc gene in rabbit hyaline chondrocytes.It will be significant to find out the scathing mechanism of interleukin-1βon the cartilage of osteoarthritis and search an effective therapy to cure it.Simultaneously, to penetrate into these problems and develop more discussion, which will bring up some new clues for the clinical therapy of osteoarthritis.MethodsThe cultured rabbit hyaline chondrocytes were separated into 4 groups at random. In the experiment, various concentrations of IL-1βwere added to the medium, and the dose in group B,C and D were 1 ng/ml, 10 ng/ml, 100 ng/ml respectively, while the group A was used to control. The cells would be affected by IL - 1βfor 12 hours. Then the effects of IL- 1βon the c-myc gene were assessed by staining, immunohistochemistry and Flow CytoMeter in the passaged cell cultures of hyaline chondrocytes, and find out whether the level of c-myc expression was significantly correlated with the dose of IL - 1β.ResultsIn the experiment, the concentrations of IL - 1βincreased by degrees, which demonstrated the level of c-myc expression was correlated with the dose of IL - 1β.The difference was significant.ConclusionIL - 1βcan cause a dose-dependent stimulation on the c-myc gene in rabbit hyaline chondrocytes and play a crucial role in osteoarthritis development by regulating chondrocyte apoptosis.
