IntroductionThe progression of carcinoma are associated with many factor, which include the factors changes of promotion vascular growth. The aim of this investigation was to study the expression of HIF-1αin vulvar squamous cell cancer,VIN,nonneoplastic epithelial disorder of skin (NNEDV)and normal skin in order to find their relation.Materials and MethodsObject :30 cases of vulvar squamous carcinoma; 35 cases of VIN (20 cases VIN I-II, 5 cases of VINIII); 40 cases of NNEDV (20 casce of squamous cell hyperplasia, 20 cases of lichen sclerosus) and 10 cases of normal vulvar skin from the First Affiliated Hospital of China Medical University pathology office. We collected the clinical material: age, clinical stage, histologic grade, nodal status.In our study,immunohistochemistry was employed with SABC method. HIF-1αpolyclonal rabbit anti-human antibody, the concentration of 1:100, CD34 polyclonal rabbit anti-human antibody, work concentration 1:80.T-test to differentiate the rates of different groups for the expression of HIF-1αin vulvar diseases and MVD were analyzed. If P < 0.05 having significant differences ,SPSS version 13.0 software was employed to analyze all data.Results1. The expression of HIF-1α The HIF-1αis not express in normal vulvar skin, 3.26±0.35 in squmous cell hyperplasia, 18.37±2.45 in lichen sclerosus, 19.81±4.32 in VIN I-II, 24.39±6.52 in VINIII and 78.25±8.76 in vulvar squamous carcinoma. There were difference between VIN I-II and VINIII (P<0.05). Between the two kinds of NNEDV, there were significant difference (P<0.05).2. The expression of MVDThe MVD in the tissues of lichen sclerosis was 10.34±2.88, squamous cell hyperplasia was 21.58±2.48, there was significant differences (P<0.05). In the tissues of VIN I-II was 21.83±3.18, there was significant differences compared with VINIII (P <0.05); MVD in vulvar squamous carcinoma was 39.88±7.65. and there was significant difference between VINIII (P <0.05).3. the relationship between MVD and HIF-1αThe MVD of 24 cases with HIF-1αpositive was significantly higher than the 6 cases of HIF-1αnegative ,suggesting that HIF-1αstimulates Neoplasia through increased angiogenesis.Conclusions1. The expression of HIF-1αis believed to play a role in the progression of VSCC. HIF-1αis related with clinical stage, histologic grade and nodal ststus.2. The HIF-1αhave nothing to do with hypodermal vasiformation of VIN.3. HIF-1αis high expression in hypoxia condition. MVD reduced in lichen sclerosus, which illustrated exist microcirculation disorder, the high expression of HIF-1αis compensation .
