| Background and objective: the etiopathogenisis of inflammatory bowel disease is not definited. in current viewpoint, cytokine may act as a importand role in this disease. Leptin ,a key hormone involved in central regulation of body weight, have great impact on immune system .In recent years,many study indicated that leptin maybe have harmful impact in UC. The objective of our study is to explore the role of Leptin and the interaction of Leptin with pro-inflammatory factors in the DSS-induced colitis model.Methods:A total of 32 BALB/C mice was randomly divided into three groups: black control group (n=10),3% DSS group (n=10) and 5% DSS group(n=12). black control group was only fed with distilled water , 3% DSS group was fed with 3 % DSS solution,5 % DSS group was fed with 5 % DSS solution. Colitis was evaluated by disease active index(DAI) and histological score, Leptin and IL-1βexpression were determined by immunohistochemical staining.Rusults: 1. DAI and histologicals core were markedly increased after the administration of 3% DSS and 5% DSS solution compared with normal control(P<0.05). 2. There were significant differences in Leptin expression among threegroups in histological samples from mice(P<0.05).The group that wasonly fed with distilled water scarcely expressed in histological samplesfrom mice.3.There was significant positive correlation of expression of Leptin andIL-1βamong the three groups.
