| Objective: To investigate the expression of COX-2 in retinoblastoma HXO-RB44 cell, and the effect of selective COX-2 inhibitor nimesulide on the growth and proliferation of HXO-RB44.To provide theoretic evidences for the application of nimesulide to the treatment of retinoblastoma.Methods: HXO-RB44 cells were cultivated in vitro. The expression of COX-2 protein in HXO-RB44 cells was detected by immunocy-tochemical (SP) method. The morphological changes of cells were observed by phase-contrast microscopy .Trypan blue dye exclusion and MTT assay were used to detect the effect on proliferation of HXO-RB44 cell after incubation for 24h,48h,72h in the different concentration of nimesulide (0,200,300,400μmol/L).Result: The positive rate of COX-2 expression in HXO-RB44 cells was 98.3%, the positive substances were expressed in the cytoplasm and the nuclear membrane. Under phase-contrast microscopy cells in each group had no apparent morphological changes. Even after 72h significant changes couldnot be seen in the 200μmol/L group. While cells with sparse density, irregular configuration, decrease of the refractive characteristic and appearance of granuliform substances in the cytoplasm could be seen in the 300μmol/L and 400μmol/L groups, respectively after 72h and 48h. The growth curve turned flatter with the increase of time and dose. From 48h to 72h the growth curve of the 400μmol/L group had a downward trend. Trypan blue dye exclusion assay revealed that the proportions of alive cells were high when the dose was low and the time was short. While with the increase of time and dose, the proportions decreased. Nimesulide could decrease A value of HXO-RB44 cells and increase the inhibitory rate in a time-and dose-dependent manner.Conclusion: 1.C0X-2 is expressed significantly in retinoblastoma HXO-RB44 cells. 2.Nimesulide has growth- and proliferation- inhibitory effects on HXO-RB44 cells in a time-and dose-dependent manner. |
