Purpose: To investigate the function of hydroxycamptothecin in influencing the apoptosis of HXO-RB44 cells in vitro and its possible mechanism.Method: Culture the HXO-RB44 cells invitro ,transfer and change the substrate .We choose five concentrations(10-5 mg/ml ,10-4 mg/ml, 10-3 mg/ml, 10-2 mg/ml, 10-1mg/ml)of HCPT and divided the cells into experimental groups and control group. The effects acted on the HXO-RB44 cells by HCPT were evaluated by MTT colorimetric assy. The apoptotic ratio and the change of the cell cycle of HXO-RB44 cells were evaluated by the flow cytometry assay. And the change of the expression levels of bcl-2 in cells was evaluated by the western blot analysisResult: The proliferations of HXO-RB44 cells in experimental groups were significantly inhibited by HCPT in different concentration (10-5 mg/ml ,10-4 mg/ml, 10-3 mg/ml, 10-2 mg/ml, 10-1mg/ml).And the inhibition ratios Were dose-dependent an time-dependent(p<0.05).The cell cycle was blocked at S phase in every experimental group. The flow cytometry tested the apoptotic peak in every experimental groups after 24h and the apoptotic rates were 5.67±1.65%, 10.09±1.03%, 12.39±0.82%, 48.54±1.27%, 18.82±0.96% respectively. There was significant difference between the experimental groups and the control group(p<0.05).western blot analysis shows that the level of bcl-2 in the cells declines significantly in each experimental group compared with control group after cultured with HCPT for 24h especially in the 10-1mg/ml and 10-2mg/ml group(p<0.05).Conclusion: HXO-RB44 cells were significantly inhibited and was block ed at the S phase of the cell cycle by HCPT in the concentration of 10-5 mg/ml to 10-2 mg/ml. This function has a dose-dependent and time-dependent manner. HCPT could induce the apoptosis of the HXO-RB44 cells in the concentration of 10-5 mg/ml to 10-1mg/ml.This is one of its anti-tumor mechanisms. HCPT could induce the apoptosis of the HXO-RB44 cells by conduct the decline of the express of bcl-2. HCPT may be use in the clinic chemotherapy of RB in some future.
