Background: In the past,disease rate of ulcerative colitis (U C)in our country was not high. Most of UC was Var. mitis or not typicalexample. Along with Elevation of industrializational level and livingstandard of the people, disease rate and sickness rate of UC haveobviously ascendent tendency. For this reason, prevention and cure of UC have important significance. It is considerd that the main morbility ofUC is to deal with heredity and dysimmunity and environment. Localimmunologic abnormality or destroy of normal immunological function isall-important role in UC. The function of cytokine (CK) in the occurrenceand development of UC had been generally acknowledged. In mainly, UCare therapied by glucocorticosteroid and salazosulfamide andimmunodepressant, with poor effect and long recover and high recurrentattacks. For the past several years people used probiotics to cure UC andgained some effect, but the precisely mechanism of action was not indetail.Objective: To comprehend the preventive and therapeutic effect inmice U C with the crude metabolic product of lactobacillus andbifidobacteria. To approach mechanism of action and to provide newthinking of prevention and cure in mice U C.Methods: We use dextran sulfate sodium (DSS) to induce BALB/cmice UC model.We usec rude metabolic product of lactobacillus andbifidobacteria to give an enema to mice and, seting up blank controlgroup and DSS model group,compare with normal sodium (NS) and salazosulfamide (SASP)to give an enema to mice. It was evaluated micestate by mice death rate, disease activity index (DAI), histological diseasescore and histological pathology transform, immunohistochemistry ofIL-10 of colon mucosa. Statistics analysis are used by t-test and partitionsof x~2 method and rank sum test (Kruskal-Wallis test).Results:1. Death rate of mice in lactobacillus group is lower than DSS modelgroup and NS group and blank control group. Its difference betweenlactobacillus group and DSS model group or NS group or blank controlgroup appeared statistical significance (P<0.05). Mice in bifidobacteriagroup have not dead, and difference of death rate of mice betweenbifidobacteria group and DSS model group or NS group or blank controlgroup appeared statistical significance (P<0.05).2. DAI of mice in lactobacillus group or bifidobacteria group islower than DSS model group and NS group. Its difference betweenlactobacillus group and DSS model group or NS group appearedstatistical significance (P<0.05). Uniformly, Its difference betweenbifidobacteria group and DSS model group or NS group appearedstatistical significance (P<0.05).3. Difference of histological disease score between eitherlactobacillus group or bifidobacteria group and DSS model group or NSgroup appeared statistical significance (P<0.05). Histological pathologytransform: In mice of lactobacillus group or bifidobacteria group,depletion of colon mucosa is few and it is scarely find mucosal erosion and bleeding and necrosis, with little inflammatory cell.4. The immunohistochemistry result of IL-10 in mice colon mucosaof lactobacillus group and bifidobacteria group are highter than DSSmodel group and NS group. Its difference between lactobacillus groupand DSS model group or NS group or blank control group appearedstatistical significance (P<0.05). Likewise, Its difference betweenbifidobacteria group and DSS model group or NS group or blank controlgroup appeared statistical significance (P<0.05).Conclusion:1. It is principal effect that the crude metabolite of lactobacillus andbifidobacteria prevent and cure mice U C.2. By promoting IL-10 express, some element of the crudemetabolite of lactobacillus and bifidobacteria educe preventive andtherapeutic effect.
