Objective: Observe the protection of chuanxiongzine on pancreatic glands, kidneys and lungs in the case of acute pancreatitis, by apply chuanxiongzine on experimental severe acute pancreatitis of SD Rat model,to approach its mechanism of action, and thus provide experimental evidence for clinical application.Methods: 90 SD rats, divided into 3 groups randomly: (1) Sham operated group (group A) :30; (2) Severe acute pancreatitis of SD Rat group (group B) :30; (3) Chuanxiongzine treatment group (group C) : 30. Blood preparation and tissue sample are harvested at 2h,6h,12h,24h respectively to detect the indexes as follows: (1) Serum amylase,BUN,CR,blood serum; (2) TNF-αand IL-6 level of serum; (3) TXB2/6keto-PGF1αlevel in blood plasma and pancreatic tissue; (4) tissue HE dyeing light microscope detection of pancreatic gland,kidneys and lungs; (5) Wet/dry of lung; (6) Mortality of three group.Results: (1)Serum amylase,BUN, CR : the level of serum amylase in group B were up-regulated notably compared to group A at 2h,peaked at 6h,and the level at different time point was significantly higher than that of group A (p<0.05); compared to group B, the level of group C down-regulated notably at 24h (p<0.05), while still higher than that of group A (p<0.05)。(3) TXB2/6keto-PGF1αlevel in blood plasma and pancreatic tissue and the relative value of TXB2/6-keto-PGF1α:the level of TXB2 were up-regulated notably in group B compared to group A since the time point of 2h, peaked at 6h,then still maintaimed on a certain high level (p<0.01); The level of TXB2 were down-regulated notably in group C compared to group B at the time point of 6,12,24h (p<0.01),while still higher than that of group A (p<0.01)。Compared to group A,the level of 6-keto-PGF1αin group B and C was higher than that of group A (p<0.01) . Compared to group B, the level of group C has no significant change (p<0.01); In contrast to group A, the relative value of TXB2/6-keto-PGF1αwere down-regulated notably in group B (p<0.01); the relative value of TXB2/6-keto-PGF1αwere down-regulated notably in group C in contrast to that of group B (p<0.01) at the time point of 6h,12h,24h, while still higher thanthat of group A (p<0.05)。(5) Observed under light microscope:①Pancreatic tissue: pancreatic tissue in group A appeared normaly,by contrast, severe pancreatic tissue cellular necrosis was seen in group B ,while compared to ingroup B, cellular necrosis appeared to be on a fairly light level in group C.②Lung tissue:extensive oedema was seen in the alveoli pulmonis and interstitial substance of group B ,there were asphyxial membrane formed in the alveoli pulmonis too ,and in the alveoli pulmonum tissue the neutrophil gathered agglomerate ;the level of cell aggregation appeared to be light in group C.③Kidney tissue: renal tubular epithelial cell necrosis was seen in group B and the dead cells ablated,so was the oedema of nephric tubules and tubular ectasia ;in group C there was only the oedema of nephric tubules,and on the light level. (6): survival rate of 3 days :Among group A,B,C ,It was 100%,16.7%,50%,respectively, survival rate of group B was notably lower than that of group C (p<0.05).Conclusion:1. Chuanxiongzine not only has therapeutic action on experimental SD rats ,but also has notably protection on kidneys and lungs besides pancreatic glands ,It has the capability of elevating the survival rate of experimental SD rats.2. It could be deduced that the possible actions of chuanxiongzine are as follows: 1. Depress the expression of TNF-α,IL-6, inhibit the effect of Inflammatory reaction, 2. Suppress the synthesis of TXA2, regulate the disequilibrium of TXA2/PGI2, lighten the effect of microcirculation disturbance.
