Objective: Analyzing different expression proteins of hippocampus of SAMP8 micetreated with traditional chinese medicine YiZhiJianNaoKeLi, this work discuss partially themechanism of YiZhiJianNaoKeLi with respect to its effectiveness to Alzheimer's disease.Method: Thirty 6-month old male SAMP8 mice are divided into three groupsrandomly: treatment group, contral group and model group.There are ten mice in everygroup.The model group mice are killed immediately after being divided into the group,thehippocampus are took out and saved with liquid nitrogen.The treatment group are drenchedwith YiZhiJianNaoKeLi concentrated fluid,the contral group mice are drenched with thesame doze of double distilled water(DDW), the two groups mice are all drenched for eightweeks, then these mice are killed and their hippocampuses are took out, subsequentlyprotein expression profiles are established with two-dimensional gel electrophoresis(2-DE),then the different expression protein spots are obtained comparing with the profiles of thethree group hippocampus through software PDQuest 7.2.Analyzing those differentexpression protein spots with matrix-assisted laser desorption/ionization time of flight massspectrometry(MALDI-TOF-MS), then obtein peptide mass fingerprinting(PMF), at last weidentify these protein spots with detabase.Result: Establishment of hippocampus 2-DE protein profiles of SAMP8 mice.Thereare thirteen more than two folds different expression protein spots identified in treatmentgroup with mass spectrometry (MS) analysis and detabase, of which eight protein spots areup-regulated and five protein spots are down-regulated.These protein spots are classified bytheir functions: related with energy generation, related with cell structure, related with sigalTransduction and hormone.Conclusion: 1. Multiple hippocampus protein spots of SAMP8 mice can be regulatedby YiZhiJianNaoKeLi, showing YiZhiJianNaoKeLi has multitarget mechanism of AD.2. Preliminary stablishment of hippocampus 2-DE protein expression profile of SAMP8mice.3.YiZhiJianNaoKeLi may improve energy metabolism and estrogen,depress neuron cellapoptosis to treat Alzheimer's disease.
