Objective: To investigate the expression levels and their relevancyof aurora A protein and Histone3 protein(H3) in normal endometrium,atypical hyperplasia(AH),and endometrial carcinoma(EC).To explorethe possible function of aurora A and H3 in pathogenesis of AH and EC.Methods: 60 cases of EC and 30 cases of AH were selected in ourstudy. 10 cases of normal endometria in hyperplasia phase were used asthe contorl. All cases were confirmed by pathology. Imunohistochemistrywas used to study the expression of aurora A protein as well as Histone3protein and estrogena eceptors(ER), followed by observation under lightmicroscopy and statistical analysis of the data.Results:1.The expression of aurora A and H3 in normal endometrium,AH,andEC increase by turns, There was significant difference between any twogroups(P<0.05). The expression of them in ECⅠwas significantlyupgraded compared to ECⅡ,ECⅢ-Ⅳand increased compared to AHⅢ, the expression of them in AHⅠwas significantly upgraded comparedto AHⅡ,AHⅢand increased compared to normal uterineendometrium. In the 60 cases of endometrial carcinoma, the expression ofAurora- A and H3 was relevant to tissue differention phase,myometrialinvasive depth and lymphatic node transfer.2.The expression of aurora A and H3 in cases which ER'sexpression are positive in EC was significantly lower than that in NEECs.3. The expression of aurora A and H3 in EC cases are positivelycorrelated.Conclusions:1. The unusual upregulation of aurora A was one of the cause forendometrium abnormal growth.2. The expression of aurora A is correlated with histological gradeand lymphatic node transfer. It might be one of events during theoccurernce of endometrial carcinoma.3. The aurora A and H3's expression are positively correlated. Itsuggests that Aurora A plays role in maglignant change though H3 pathway.4. The abnormal high expression of aurora A and H3 in the NEECsmay give new direction to investigate the pathogenesis, treatment andprognosis of NEECs.
