Soyisoflavones is main bio-active component in soybean, whichpossess of the properties such as cholesterol lowering, syndromecarcinogenesis prevention, alleviation of osteoporosis menopause and soon. They have been be Widely used in pharmacy, healthful foods anddietary supplements. In this paper, either the isolation method ofsoyisoflavones from soybean extract was studied or the purificationmethod.1,A method was developed for isolation of four major soyisoflavones from soybean extract by the HPD-100 macoporous resins.Ethanol aqueous solutions with various concentrations were applied toelute four objective isoflavones under a gradient elution mode. Thecontent of daidzin, genistin, daidzein and genistein was 49.31, 60.38,19.33 and 8.14% in each product, respectively. The method can beapplied to pre-purify four major soybean isoflavones.2,A method was developed for hydrolysis of soybean isoflavones bythe microwave in order to get the aglycones. The hydrolyzing conditionwas studied and optimized. Compare with the traditional method, refluxhydrolysis, microwave hydrolysis is simpler, faster and more efficient.The method can be applied to quickly yielding the isoflavone aglycones.3,A method was developed for preparation of Daidzein and Genistein bypreparative HPLC. The conditions of preparative HPLC was optimized by comparing the proportion of organic phase in mobile phase, flow rate,sample volume. The method was proved to be a simple, convenient andeffective for produce high purity Daizein and Genistein. The purityproduct can be the reference substances in the research of TraditionalChinese Medicine.4,A method was developed for isolation of soybean isoflavonesaglycone and glycoside only in the medium of water. The extractionconditions were studied and optimized such as kind of solvent,temperature, time of extraction procedure and the ratio of material vs.volumne. Result showed that content of glycoside in supernatant productwas high to 32.24% and aglycone was only to 3.05%. On the contrarythey were 4.28% and 72.03% in the deposit, respectively. The method canbe applied to produce soybean isoflavones aglycone and glycosideindustrially for its good property of simple cheap and nontoxic.
