Alzheimer's disease , senile dementia ,is one kind of declined central nervous system disease, at present ,we think that the neurotoxicity ofβ-amyloid protein deposits in the brain is the criticality reason of AD.Clinically,we use estrogen replacement therapy to cure AD,the curative effect is fairly good,but because of the latent cancerogenic effect ,estrogen's clinical use is constrainted. Soybean isoflavone is one kind of phytoestrogen,which has has the estrogen effect ,and has the health care effect to human being . Soybean isoflavone has no notable toxicity plant to human being too.We can use Soybean Isoflavone to exploit a ideal medicine to replace estrogen.Reserarchs show that Soybean contains many Isoflavone .We know that Isoflavone Aglycone has stronger activity than Isoflavone and is easier to admit for the human body.In this ,we study the prepare techniques of high purity Soybean Isoflavone Activity Aglycone Component from Soybean Isoflavone coarse extraction and study its protection effect of nerve cell.【Objective】To extract and purify Soybean Isoflavone Activity Aglycone Component from Soybean Isoflavone coarse extraction and Determine contents of Activity Aglycone by HPLC ;To study its protection of PC12 cell against oxidative damage inducedby H2O2 and neurotoxicity by Aβ.【Methods】 (1)We use acid hydrolysis to transformat glucoside of Soybean Isoflavone coarse extraction into Activity Aglycone and recrystallisation to improve contents of Soybean Isoflavone Activity Aglycone Component(2)We PC12 cell impaired model induced by H2O2 and Aβ.To study Soybean Isoflavone Activity Aglycone Component protection of PC12 cell against oxidative damage inducedby H2O2 and neurotoxicity by Aβ, We Observe under the microscope , use MTT to evaluate the survival rate and LDH to check the LDH activity.【Results】(1) We optimize the prepare techniques of Soybean Isoflavone Activity Aglycone Component and contents of three Activity Aglycone are 71%,which is Determined by HPLC.Comparede with Soybean Isoflavone coarse extraction, the contents increase by 10.36 times , the yield is 45.1%.(2) After being treated byH2O2 (150umol/L) and Aβ25-35(20umol/L) ,PC12 cell is strongly damaged .The PC12 cell form is abnormaland the quantity of cell is notably decreased.Compare to the normal control ,the the survival rate obviously reduces(P <0.05) and the LDH activity rise.The form of PC12 cells, which were protected by Soybean Isoflavone Activity Aglycone Component ,is normal ; the the survival rate of cell is near normal control and the LDH activity is obviously lower than damaged group (P <0.05).【Conclusion】Our studies show that Soybean Isoflavone Activity Aglycone Component can obviously improve the abnormal form of PC12 cell induced by H2O2 and Aβand show a good protection of PC12 cell against oxidative damage inducedby H2O2 and neurotoxicity by Aβ.The result provided a basis for the deep research of Molecular mechanism and developding a clinical medicine.
