Objective An amoxicillin resistance strain was obtained by inducing in vitro,The function of spots expressed differentially were identified.Method Amoxicillin resistance strain was selected from the strain J99 by serial passage technique in vitro.One amoxicillin-resistance(AMX~r)strain was obtained.Proteins samples were prepared by precipitation with trichloroacetic acid(TCA)in acetone,and separated by two-dimensional electrophoresis(2-DE).Spots expressed differentially in-gel digested and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS).Results Then we compared the maps of the primary and the resistant strains and identified the different-quantity proteins with MALDI-TOF and database searching.The result indicated that eight protein spots were obvious different in the protein profiles of J99 and the AMX~r strain. One spot of S-adenosylmethionine synthetase was observed only in the primary strain,One spot of co-chaperonin GroESwas observed only in the resistant strains.Two spots of 30S ribosomal protein S4 and flagellin were up-regulated in the resistant strain.Four spots of 2-oxoglutarate ferredoxin oxidoreductase,3-OXOACID COA-TRANSFERASE,SUBUNIT A,NADH dehydrogenase subunitâ… and hypothetical protein were down-regulated in the resistant strains.Conclusions An AMX~r strain was obtained.The bacteria was effected through the valid medicine,the metabolism of bacteria generate change.The expression of protein was different, The study of the discrepant proteins can understand mechanism of Helicobacter pylori resistant to AMX.
