The Influence Of Propofol To The Tumor Necrosis Factor-α Induced Apoptosis Of Vascular Endothelial Cell And The Activity Of Caspase-8

Objective The aim of the study was to observe the morphology change of the tumor necrosis factor-αinduced apoptosis of vascular endothelial cell and the influence of the different density of Propofol to the apoptosis rate of vascular endothelial cell and the expression of caspase-8.Methods Human umbilical vein endothelial cell(HUVEC)were inoculated into six-holes cultivation board.This experiment includs six groups:(Using P to express the density of Propofol,and the unit isμmol/L)Group A(P0)Group B(P25)Group C(P0+TNF-α)Group D(P25+TNF-α)Group E(P50+TNF-α)Group F(P100+TNF-α)Adding different density Propofol into corresponding groups,incubate thirty minutes in incubator(37℃,5%CO2),then Adding TNF-αinto corresponding groups which density is 2000U/ml in the end,keep them at incubator 24 hours.To detect the apoptotic index by Flow Cytometry and the expression of caspase-8 by immunohisto-chemistry.Result The apoptosis rate and the expression of caspase-8 of all groups:Group C(P0+TNF-α)is higher than the other five groups obviously(P＜0.05);Group D(P25+TNF-α)is lower than Group C(P0+TNF-α)(P＜0.05),but is higher than the other four groups(P＜0.05)Group E(P50+TNF-α)is lower than Group C(P0+TNF-α)and Group D(P25+TNF-α)(P＜0.05),but is higher than the other there groups(P＜0.05)Group F(P100+TNF-α)is lower than Group E(P50+TNF-α),Group D(P25+TNF-α)and Group C(P0+TNF-α)(P＜0.05),but is higher than Group A(P0)and Group B(P25)(P＜0.05)The difference between Group A(P0)and Group B(P25)have no statistics meaning(P＞0.05).Conclution1)TNF-αcan lead to the apoptosis of vascular endothelial cell,2)The expression of caspase-8 heighten at the apoptosis of vascular endothelial cell,3)Propofol can inhibit the the apoptosis of vascular endothelial cell and the expression of caspase-8,4)The protection of Propofol to the apoptosis of vascular endothelial cell and inbibiton the expression of caspase-8 is dose-dependent.
