Objective:To investigate the effect of high glucose on the production of nitric oxide(NO) and Intercellular adhesion molecule-1(ICAM-1)in human umbilical vein endothelial cell (HUVEC)in different times,and to further study the intervention of rosiglitazone.To illuminate the potential protection of rosiglitazone on vascular system.Methods:Human umbilical vein endothelial cell was cultured in different dulbecco's modified eagle medium(DMEM)different times(0 hour,24 hours,48 hours,72 hours,96 hours).The experiment was divided into three groups randomly:Control group(concentration of glucose:5.5 mmol/L),high glucose group(concentration of glucose:25 mmol/L),and rosiglitazone group(concentration of glucose:25 mmol/L,concentration of rosiglitazone:105M). NO level,reflected by NO3-+NO2-in supernatant,was detected by HNO3 reductance assay. Meanwhile ICAM-1 in supernatant was detected by ABC-ELISA.Results:In high glucose group,comparing with control group,the concentration of NO was significantly increased at the time of 24 hours and then down as cultured time goes by.It was significantly decreased at the point of 48 hours and then down to base line at 72 hours. Meanwhile,in rosiglitazone group,comparing with high glucose group,NO level was significantly decreased at the time of 24 hours.But rosiglitazone increased NO level significantly at 48 hours,72 hours and 96 hours comparing with high glucose group;In addition, comparing with control group,ICAM-1 level in high glucose group was found to be significantly increased at all time points and took on time-dependence.Rosiglitazone could decrease the content of ICAM-1 to normal level.Conclusion:High glucose can damage endothelial cell function.High glucose can increase NO level at first then decrease it.On the other hand,high glucose can increase ICAM-1 level. Rosiglitazone could reverse the content of NO and ICAM-1 to normal level.Rosiglitazone can protect blood vessel endothelium function.and provide a new method to prevention and cure diabetic vascular complication effectively.
