Stimulation Of VEGF In Rabbit Phallus With Hypospadias: Role Of HCG

Objective To research the etiopathogenisis of hypospadias and the significance of Vascular endothelial cell growth factor(VEGF)in rabbit phallus with hypospadias using human chorionic gonadotrophin(HCG).Methods To select forty pregnant rabbits(New Zealand species),from the 19th day of their pregnancy,received gavage of Finasteride Tablets(PROSCAR)for 0days(group A),4 days (group B),5 days(group C),6 days(group D),7 days(group E)which begin at GD(gestation days)in a dosage of 10mg·kg^-1·d^-1;after withdrawal,the pregnant rabbit ran a spontaneous labour; from on the35st day after birth the male whelps were observed on the morphological change of their external genital processes.Congenital hypospadias in rabbit(New Zealand space)were established.All of rabbits were divided into the group with normal sodium of intramuscular injection and the group with HCG of intramuscular injection,the group with HCG of intramuscular injection were give randomly into 4 groups with HCG of intramuscular injection for 7 consecutive days in a dosage of 100IU,200IU,400IU and 600IU respectiveiy.To detect concentration of VEGF in rabbit phallus with control group and test group using ELISA respectively.Results Male whelps were seen in group A(0),group B(22.2%),group C(85.2%),group D(95.5%),group E(100%),and their urethral meatus were present on the ventral side of the penis.To detect concentration of VEGF in rabbit phallus with control group and test group using ELISA respectively.The total concentration of VEGF in rabbit phallus with hypospadias was lower than these of control group(P＜0.05).It was distinction between normal group and with normal sodium of intramuscular injection of rabbit with hypospadias group(P＜0.05),and between normal group and with HCG of intramuscular injection in a dosage of 100IU,200IU and 400IU (P＜0.05).It was not distinction between normal group and with HCG ofintramuscular injection in a dosage of 600IU(P＞0.05).Conclusions Congenital hypospadias in rabbit(New Zealand space)were established using pregnancy rabbit,given a Finasteride Tablets in a dosage of 10 mg·kg^-1·d^-1for consecutive from 5 to 7 days.The total concentration of VEGF in rabbit phallus with hypospadias is decreased, which may be related to the genesis of hypospadias.Exogenous of HCG could stimulate the increase of concentration of VEGF in rabbit phallus with hypospadias in a dosage.
