Objective To assess the influence of vascular endothelial growth factor(VEGF) suppressed by vector-based small interfering RNA(siRNA)on the expression of vascular endothelial growth factor-C(VEGF-C)and its receptor(vascular endothelial growth factor receptor-3,VEGFR-3)in human tongue squamous cell carcinoma cell line(Tca8113).Methods Two siRNA targeting VEGF eukaryotic expression vector (Pu-VEGF-siRNA1,Pu-VEGF-siRNA2),and eukaryotic expression vector which was used as experimental control,were transfected into Tca8113 cells by Lipofectamine2000,respectively.Non-transfected cells were used as negative control.VEGF,VEGF-C and VEGFR-3 mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR),respectively.Results Compared with experimental and negative controls,the expression of VEGF mRNA were significantly decreased in the Pu-VEGF-siRNA1 group and Pu-VEGF-siRNA2 group(P<0.05);however,the expression of VEGF-C mRNA(P=0.845)and VEGFR-3 mRNA(P=0.980)were not significantly decreased in Pu-VEGF-siRNA1 group and Pu-VEGF-siRNA2 group.Conclusions Vector-based siRNAs targeting VEGF can efficiently down-regulate VEGF expression in Tca8113 cell in vitro.But the down-regulated VEGF can not influence on the expression of VEGF-C and VEGFR-3 mRNA.
