Effects Of Sibelium On C-fos Expression Within Trigeminal Nucleus Caudalis Of The Rat Following Glyceryltrinitrate Infusion

Objective:To observe the influence of sibelium on c-fos expression within trigeminal nucleus caudalis of the rat following10mg/kg glyceryltrinitrate infusion.Methods:The experimental migraine rats were randomly divided into four groups:Sham group (GroupⅠ,n=6),Control group(GroupⅡ,n=6),,Treatment group(GroupⅢ,n=6)and Preventment group(GroupⅣ,n=6).10mg/kg glyceryltrinitrate infusion to copy migraine animal model. According to reference,1.04ml/kg physiological saline infusion to Sham group(GroupⅠ,n=6). 10mg/kg glyceryltrinitrate infusion to copy migraine animal model in Control group(GroupⅡ,n=6),Treatment group(GroupⅢ,n=6)and Preventment group(GroupⅣ,n=6).In 1.04ml/kg distilled water infuse to stomach control group after copying model 30min.1.04mg/kg sibelium infuse to stomach control group after copying model 15-20min.1.04mg/kg sibelium infuse to stomach control group before copying model a week.4h after copying migraine animal model, 1%50mg/kg Pentobarbital to paralyze rat,which 4mm trigeminal nucleus caudalis(TNC) removed.The expression of c-fos immuno-reactivity was detected with immunohistochemical method.Results:1):90.9%of copying migraine animal model in 22 rat.The GTN rats appeared reddish two ears,head shaking,scratching head with forepaws,activity increasing in 5～10min, peaked in 40～60min,keeped with 1～3h.The sham rats appeared reddish two ears,head shaking, scratching head with forepaws,activity increasing small.Treatment group and preventment group decreased at 60min,disappeared at 120min.2):In OLYMPUS,the cell was tasteless in background,the positive cell was yellow in TNC.The positive cell in control group was obviously increased between treatment group and preventment group.3):Different groups were significant difference among four groups(F=20.365,P＜0.01).There were significant difference comparing with other 3group(P＜0.01).Sham group,treatment group and preventment group were no ignificant difference(P＞0.05).Conclusion:1):The model is a good one which can used for evaluating drug' s effect and studing pathogenesis.2):The experimental result support trigeminovasular.3):The result suggested that sibelium could change c-fos expression,it could be used to prevent and treat migraine.
