Objective:To investigate the effect of Retinoic acid on proliferation and TGF-βsecretion of Human mesangial cells treated with high concentration of glucose.Methods:Mesanngial cells were incubated with different concentrations of Retinoic acid under high glucose condition.Cell proliferation was assessed by MTT assay.TGF-βsecretion was detected By ELISA.Results:High glucose(30mmol/l)could induce mesangial cell proliferation.It got to the best at 48hours(P<0.05).Ranging from 10-9- 10-6mmoL/L retinoic acid can inhibit the proliferation of mesangial cells under high glucose concentra-tion and showed dose- dependence (P<0.05).High glucose can results in TGF-βsecretion of human mesangial cells.Retinoic acid also decreases TGF-βsecretion of mesangial cells under high glucose condition and showed dose- dependence(P<0.05).Conclusion:High glucose could induce mesangial cell proliferation and TGF-βsecretion. AtRA can inhibit high glucose-induced proliferation of mesangial cells and TGF-βsecretion.
