Objective:Diabetic nephropathy(DN) is one of the common chronic complications of diabetes mellitus and the major cause of end-stage renal disease (ESRD). The early pathological changes of DN include mesangial cells (MCs) increased, extracellular matrix (ECM) accumulated and basement membrane thickened. The better ways which we must research for is to prevent progress. However,the recent study has demonstrated that inflammation is an important cause in chronic progressive renal disease in diabetic patients.The studies demonstrated that ATRA had renoprotective effect on some renal diseases animal modeles throught an anti-inflammatory activity. Mesangial cells (MCs) has all subtypes of retinoic acid receptor family.So I will study whether ATRA protect DN.MethodscRat MCs were cultured with low and high glucose and 10-8,10-7 or 10-6mol/L of ATRA,Cell proliferation was tested with MTT and observed by spectrometry 12,24,48 and 72h later.The concentration of type IV collagen in supernatant was measured by ELISA.Results:1.The MCs prolifeation presented a time-dependent feature in high glucose.The high glucose inhibit proliferation on 72h.ATRA could inhibit the high glucose induced proliferation in a time-and dose-dependent feature.2.The concentration of type IV collagen in supernatant with high glucose was higher than that of control group;High glucose plus ATRA group had ao lower concentration of type IV collagen than that of high glucose group.Conclusion:1. High concentration of glucose can stimulate MCs proliferation with a time-dependent feature. ATRA can inhibit such a high glucose-induecd proliferation of MCs also in a time-and dose-dependent way.2.High glucose can increase the concentration of type IV collagen in supernatant with a time-and dose-dependent feature. ATRA can decrease type IV collagen also in a time-and dose-dependent way.3. ATRA protect early DN.
