Objective: In order to reveal the mechanisms of rh-endostatin, the suppression effects ofrh-endostatin on HUVECs in different activating levels were analyzed. Methods: Toprepare three kind of HUVEC. To detect the marker of CD146,CD105,CD62E on thesurface of adherenting HUVEC,exponential phase of growth HUVEC, activated withTNF-alpha HUVEV and the alteration by flow cytometry. The effects of rh-endostatin onthese three kinds of HUVEC were indicated by the variation of the expression of CD105and CD62E on the cells. The relationship between the change of cell markers and the timeand the concentration of rh-endostatin were analyzed by SPSS 11.5.Results:1,Thepercentage of cell marker before treatment with rh-endostatin: (1) Before logarithmicgrowth phase: The percentage of CD105~+ cells increased with a time-dependent style.However, the prevalence of CD62E~+ cells has no change. (2) Logarithmic growth phase:The percentage of CD146~+ CD105~+ cells remains in high level, while the prevalence ofCD62E~+ cells is low. (3)HUVEC activated with TNF-alpha: CD146~+ cells remains in highlevel. The prevalence of CD105~+ cells decreased as dose-dependent style in 6 hours, whileit increased to the level of the cells in logarithmic growth phase after 24 hours. Thepercentage of CD62E~+ cells increased as dose-dependent style in 6 hours, which reachedthc peak amplitude at 250IU/ml and decreased after 24 hour. 2,The variation of cellmarkers expression: (1) The change of CD105: The expression of CD105 was correlatedwith the time and dose of rh-endostatin in all group. (2) The change of CD62E: Theexpression of CD62E was correlated with the dose of rh-endostatin in the group ofstimulating with TNF-αin 24h. Conelusions: 1,CD146 expressed on all kinds ofendothelial culls, including "resting" and "activating" cells. There is no influence of TNF-α on the expression of CD146.2,CD105 has been regarded as a specific marker of theendothelial cells in early-stage of activation because it is expressed on the endothelial cellswith proliferating and adherent activities.3,CD62E is a marker of the endothelial cells inlate-stage of activation. which increased by TNF-α.4,Definite dose rh-endostatin inhibitedthe activation of endothelial cells which indicated by the expression of the markers ofactivated of endothelial cells. However. rh-endostatin did not induce the apoptosis of thecells.
