The Protection Of Four Herbal Compound Formulae On Stress-induced Tissue Injury

Objective: We observed the influence of Chaiyu Decoction, Shaogan Decoction,Jinlingzi powder and Tongxie prescription on different animal models ofpsychological stress and investigated the protective effect of these four traditionalChinese medicine compound prescriptions on stress-induced tissue injury, andprovide experimental basis for clinical use.Method: Part 1(ineluding experiment 1 and 2) To observe the effects of four herbalcompound formulae on acute psychological stress mice. The model was produced byrestraint-immersion. This kind of stress maybe causes gastric ulcer and hepatic injury.The specific methods are as follows: 150 healthy KunMing mice were divided into 15groups, namely normal control group, model group, Chaiyu Decoction groups of low,middle and high dose, Shaogan low, middle and high dose, Jinlingzi powder low,middle and high dose, Tongxie prescription groups of low, middle and high dose andpositive control group(ranitidine+Vit E). The experiment lasted for 7 days. For thefirst five days, mice in various groups were respectively fed with each drugs once aday, except normal control group. Mice in model group were administered withnormal saline. Fasting 24 hours after the last administration, then put the mice intothe water. The water surface should reach the xiphoid of the mice and the temperatureof water was 18℃±1℃. The blood samples were taken 20 hours later and serumswere separated. Then the stomach and the liver were dissected. Measured the ulcerindex in the stomach, and determined the content of MDA in the liver and ALT in theserum.Part 2(including experiment 3 and 4) To observe the effects of four herbalcompound formulae on chronic psychological stress rats and investigated theinfluence of these four drugs in endocrine system, immune functions and the brainmonoamine transmitter. This kind of stress maybe causes major depression. The specific methods are as follows:120 healthy SD rats were divided into 15 groups,namely normal control group, model group, Chaiyu Decoction groups of low, middleand high dose, Shaogan Decoction groups of middle and high dose, Jinlingzi powdergroups of middle and high dose and Tongxie prescription groups of low, middle andhigh dose and three positive control groups(clomipramine hydrochloride, sertralineand maprotiline). The experiment lasted for 28 days. For the first 14 days, we use 10irritant factors stimulate the rats once a day. This 10 factors including fasting 24 hours,water-depriving 24 hours, moist pad and inclining feeding on a slope, bounding 2hours,10 min in ice water,10 min in hot water, strange odor, strange objects,alternating the day and night and electric shock. From the 15th day, rats in variousgroups were respectively fed with each drugs once a day, except normal controlgroup. Mice in model group were administered with physiologic saline. Fasting 24hours after the last administration, then isolated the brain and the liver. Measured thecontent of cortisol and IL-2 in the serums, and 5-HT in the brain.Result: Experiment 1. Four herbal compound formulae can reduce the ulcer indexand increase ulcer inhibiting rate.Experiment 2. Four herbal compound formulae can reduce the content of ALT inserums. And there were no significant difference. Four herbal compound formulaecan reduce the content of MDA in liver. But there were significant difference betweenChaiyu Decoction and Jinlingzi powder.And the same as Jinlingzi powder andTongxie prescription.Experiment 3. Four herbal compound formulae can improve the behavior of the rats.They can enhance the content of IL-2 in serums. But the influence on Cortisol isdifferent. All kinds of drugs in this experiment can reduce the content of Cortisol inblood serums except Chaiyu Decoction and clomipramine.Experiment 4. Four herbal compound formulae can increase the content of 5-HT in hippocamp except Tongxie prescription. And there were no significant differenceamong these herbal compound formulae.Conclusion: Chaiyu Decoction, Shaogan Decoction, Jinlingzi powder and Tongxieprescription have the protect effects of stress damage on different organs, includingthat four herbal compound formulae have the protect effects on acute psychologicalstress induced gastric ulcer and hepatic injury. The functional mechanism maybecorrelated with the decrease of damage of lipid peroxidation. They can also inducethe content of IL-2 in blood serums.in order to enhance the immunity.
