ObjectiveToexplor the expression and significance of the apoptosis related protein Gapase-3 and Bcl-2, NF-kB genes in alcoholic liver disease rats.Methods1. The Model of Alcoholic liver Disease: Rats were randomly divided into model group (n=48) and control group (n=48).The dose of 40% ethanol (8g/kgbody/weight) was administerd by gastric gavage twice a daily for 12 weeks in the model group, and the control group was received an equivalent amount of saline by gavage. The rats were killed at the end of 8W and 12W. The blood was taken from the left ventricle.2. The activities of serum ALT and AST were detected by autobiochemistry instrument.3. The Liver tissues were fixed by 10% formalin, paraffin embedded and cut 5μm thick sections. The pathological changes and proliferation degree of collagenous fiber were observed under light microscope after H-E and Sirius-red staining.4. The hapatocytes apoptosis was evaluated by the TUNEL assay in liver tissue. Expression of Caspase-3, Bcl-2 and XF-kB were determined by Immunohistochemical SABC method.Results1. Compared with that in the control group, the level of serum ALT,A ST was signilicantly increased in the model group and ratio of ALT and AST was large than two (P<0.05).2. Compared with that in the control group, the hepatocytes hecame swe lling and macro and microvesiculal steatosis, spotty and focal necros is, inflammation were observed in model group, and pathological changes was gradually progression.3. Apoptotic hepatocytes were prominently aggregate in and around of the necrotic area in model groups. The labeling index of hepatocytes apoptosis was significantly higher than in normal control group (P<0.05).4. The positire cells of Caspase-3, Bcl-2 and XF-kB were observed around central vein and necrosis area in model group, and the expression intensity of the apoptosis related protein Caspase-3 and Bcl-2, XF-kB genes was significantly higher than that of normal control group(P<0.05).ConclusionOccurrence of alcoholic liver disease was closely associated with heaptocytes apoptosis. The Caspase-3, Bcl-2 and XF-kB partaked in occurrence and progress of alcoholic liver disease.
