Subject: To research the effects of maize plumple extracts on the female reproductive organs and the mammary epithelium in vitro.Method:①Produce mice aged model with D-gal, mensurate the green weight and apparatus index of ovary and uterus, SOD, GSH-Px, MDA and total protein content in the tissue homogenate and estradiol in serum.②Culture the mammary epithelium in vitro and identify it by using cytokeratin-18 monoclonal antibody and casein mRNA reverse transcription.③Add different concentration of maize plumule extracts to the cell culture medium, mensurate the cell curve of growth, doubling time, divisional index and total protein content.④Culture the cell in serum-starved medium and add different concentration of maize plumule extracts, mensurate the cell proliferation, cell breeding ratio and degree for protection; determine the bcl-2 gene expression.⑤Culture the cell in serum-starved medium and add different concentration of maize plumule extracts, then determine the cell total protein content.Results: 1, The results of the female mice reproductive organs texts show that:①The contents of SOD, GSH-Px, total protein and E2 in the mice injected D-gal are declined (P<0.01) and the MDA content are increased ((P<0.01) conpared with the nomal.②The two concentrations of EMP can promote the contents of SOD, GSH-Px and total proteins(P<0.01, P<0.05), reduce the MDA content(P<0.01).③EMP can increase the E2 content in serum, the value are (6.68±0.66)and(6.66±0.56)respectively, and have the significance conpared with the normal((P<0.05). 2, The results of the mammary epithelium cultured in vitro show that:①20 mg/L concentration of EMP can shorten the cell doubling time (P<0.05) and increase the cell divisional index (P<0.05), and have the significant difference;②The EMP can not induce the casein expression;③The cell proliferation is reduced under the serum-starved condition and 20 mg/L and 40 mg/L EMP have the postponable effect (P<0.05), and have the significant protective effect(P<0.05).④The mammary epithelium can apoptosis under the serum-starved condition, and the bcl-2 gene expresses. 20 mg/L and 40 mg/L concentration of EMP can induce the bcl-2 expression, and the average optical density are (0.3777±0.0111) and (0.4163±0.0134) respectively, and have the significant different (P<0.05, P<0.01) compared with the serum-starved one. The 10 mg/L concentration are not significant.⑤20 mg/L and 40 mg/L concentration of EMP can significant increase the total protein content(P<0.05), and the 10 mg/L concentration are not significant.Conclusion: EMP have the significant effects on aged female reproductive organs. It can improve the mammary epithelium to proliferate, and also can prevent the apoptosis of the cell.
